Objectives:Investigating the proliferation inhibition of cucurmosin(CUS) combined with the commonly used chemotherapy drugs of pancreatic cancer, like Gmcitabine(GEM),Fluorouracil(5-FU),Paclitaxel(PTX) and Cisplatin(DDP),on human pancreatic cancer cell line Bx PC-3,which could provide some theoretical foundation and experimental basis for pancreatic cancer clinical application of CUS combined with some chemotherapy drugs.Methods:1.Sulforhodamine B(SRB) assay was used to detect the proliferation inhibition of CUS on human pancreatic cancer cell line Bx PC-3 in vitro. 2. SRB assay was used to detect proliferation inhibition in human pancreatic cancer cell lines Bx PC-3 in vitro after the treatment of CUS combined respectively with GEM,5-FU,PTX or DDP.3.Colony forming assay was used to observe the colony forming efficiency in Bx PC-3 cells treated by CUS combined respectively with GEM,5-FU,PTX or DDP. Jin’s formula was used to assess the synergistic effect of these combinations.Result:1.Exposure to CUS at 0.015625,0.03125,0.0625,0.125,0.25,0.5 and 1.0μmol/L for 24,48 and 72 h,the proliferation inhibition effect was elevated gradually with the increase of concentration and extension of time.At 24,48 and 72 h after CUS treatment,the half maximal inhibitory concentration(IC50) were(0.696±0.083)μmol/L,(0.357±0.036)μmol/L and(0.216±0.016)μmol/L,which imply that CUS inhibited Bx PC-3 cells’ growth in a time and dose dependent manner. 2. Except the few concentrations of combinations that high concentration of CUS combined with high concentration of these chemotherapy drugs,the proliferation inhibition rate of CUS combined with GEM,5-FU,PTX or DDP on Bx PC-3 cells were all higher than each single drug alone,q>0.85,and the combined effects were more obvious at low concentrations.3.The colony formation inhibition rate of the combined treatment groups were all higher than each single drug alone,q>1.15.Conclusion:The proliferation inhibition of Bx PC-3 cell was remarkly inhibited by CUS alone,and CUS combined with GEM,5-FU,PTX or DDP show a good synergistic effect on growth inhibition in Bx PC-3 cells in vitro.
