Objective: To study the protective effects on mice with acute irradiation damage and mechanism of bicyclol. Methods: 24 BALB/C mice were randomly divided into 4 groups: blank control group, merely irradiated group, bicyclol+irradiated group, merely bicyclol group, each group of 6 mice. From 3 days prior to the irradiation day, bicyclol+irradiated group and merely bicyclol group were given 1 times per day to the mice of bicyclol by intragastric administration for 3 d continuously, blank control group and merely irradiated group were given physiological saline instead. After being intragastric administration on the 3th d, merely irradiated group and bicyclol+irradiated group were given chest and epigastric a 6MV-X-ray single-wide irradiation of 12 Gy. The mice were continued to be intragastric administration for 3 d after radiation. And then the mice were put to death at the 28 th d. The protective effect were observed from several aspects as follows(1)the general conditions: Record the weight, mental states, dietary status and activity condition of each group mice after radiation;(2)oxidative stress index: Detect the content of MDA and the activities of SOD, GSH-Px of liver, lung tissue in each group mice at the 28 th d after radiation;(3)pathohistological examination: The pathology change of liver, lung were observed with HE staining.Results:(1)the general conditions: The mice showed slow growth, depressed, reduc ing diet and slow in reacting.(2) oxidative stress index: ①Compared to the blan k control group, MDA content was significantly increased(P<0.01), while SOD a nd GSH-Px activity were significantly reduced(P<0.01) of liver, lung tissue in t he merely irradiated group and bicyclol+irradiated group mice;②Compared to th e merely irradiated group, MDA content was significantly decreased(P<0.01), wh ile SOD and GSH-Px activity were significantly induced(P<0.01) of liver, lung t issue in the bicyclol+irradiated group mice; ③Compared to the blank control gr oup, MDA content, SOD and GSH-Px activity showed no significant difference(P>0.05) of liver, lung tissue in the merely bicyclol group mice.(3)pathohistological examination: Compared to the blank control group, the liver, l ung tissue showed obvious inflammation in the merely irradiated group and bicyc lol+irradiated group mice; Compared to the merely irradiated group, the liver, lun g tissue showed lightened inflammation in the bicyclol+irradiated group mice; The re were no obvious changes in the liver, lung tissue in the blank control group merely bicyclol group mice. Conclusion: Bicyclol can protect the acute radiation-injured mice, whose mechani sm of action may be relevant to lighten lipid peroxidation by free radical...
