Objective:(1) The research group culture BMSCs from New Zealand White rabbits,trying to explore and improve the systems approach of separation,extraction and identification of BMSCsMVs;(2) To investigate BMSCs-MV’s effect on chondrocyte’s proliferation by co-culturing BMSCs-MVs with chondrocytes;(3) To observe BMSCs-MV’s effect on chondrocyte extracellular matrix synthesis activity by co-culturing BMSCs-MVs with chondrocytes;Methods:(1) Isolation,culture and identification of BMSCs from 2-month-old New Zealand rabbits;(2) Isolation and identification of BMSCs-MVs;(3) Obtain chondrocytes from 3-month-old New Zealand white rabbits;(4) Co-culturing BMSCs-MVs with P3 chondrocytes, chondrocyte’s proliferation activity was tested by CCK-8 and Ed U; RT-PCR was used to detect chondrocyte extracellular matrix synthesis activity;Results:(1) CCK-8 and Ed U assay illustrated BMSCs-MVs can promote chondrocytes proliferation(P<0.01); the proliferation activity promoted with the increase of BMSCs-MV’s dose(P< 0.01);(2) RT-PCR showed the m RNA expression of Agg and type-Ⅱcollagen increased after co-culturing chondrocytes with BMSCs-MVs; the m RNA expression of type-Ⅹcollagen, MMP-13 and TIMP-1 decreased.Conclusions:(1) BMSCs-MVs can promote chondrocyte’s proliferation;(2) BMSCs-MVs may protect cartilage by increasing the m RNA expression of Agg and type-Ⅱcollagen and decreasing that of type-Ⅹcollagen,MMP-13 and TIMP-1;...
