Objective:To use the high-resolution melting technology analysis laryngeal squamous cell carcinoma patients with UGT1A1 genetic polymorphism locus;Study the associated with UGT1A1 genetic polymorphisms and the risk of attacked by laryngeal cancer, to screening out the possible bio-marker gene locus for diagnosis laryngeal cancer as earlier.Methods:Experimental treatment group were collected in January 2014 to February 2015 during the Medical University of Shanxi first Hospital, Shanxi Cancer Hospital,Department of ENT(ear-nose-throat) with Head and Neck Surgery in patients with laryngeal cancer of 103 cases. All patients were confirmed by endoscopic pathology diagnosis of laryngeal squamous cell carcinoma. According to the growth of the scope and severity of the tumor invasion, according to the 2002 edition of the International Anti-Cancer Association(UICC) TNM standards are divided into: 0 2 cases, I in 30 cases,II in 38 cases. Stage III 27 cases of stage IV 16 cases. In the control group were 220 cases in the medical center’s physical health examine. Inclusion of all people in the control group were excluded from the throat cancer diseases and various kinds of tumor. Each sample of two groups all were collected blood samples in the early morning with empty stomach,then abstract white cell,s DNA, and put the DNA into the freezer and stored at-20 ℃. Selection of four kinds of samples in the experimental group and the control group by random stochastic, PCR amplification, pass by means of PCR amplification products to choose the right temperature. HRM read out the results pass by means of the peak of every curve,approximately classified into three different kinds of curves, each kind of curve selected 4cases randomly, send sequenced to make sure of the results of HRM and further statistical analysis. Comparative Analysis two groups,and find out the differences about genetic polymorphisms, and the use of a relational database for the likely locus bio-marker detection predict.Results:1.The difference between patients with laryngeal squamous cell carcinoma sites UGT1A1 rs4148323 polymorphism and the control group of healthy people was statistically significant(P <0.05, OR = 2.61,95% CI = 1.42-3.77).2. Carrying G genotype than the non-carrying G allele, and contain smoking, the riskof suffering from drinking history laryngeal squamous cell carcinoma increased(P<0.05).3. UGT1A1 gene rs4148323 locus with smoking, alcohol interaction effect is moresignificant(Smoking: OR = 6.48,95% CI = 2.89-9.14; Drinking: OR = 4.55,95% CI =2.83-6.24).Conclusions:UGT1A1 rs4148323 gene polymorphism [A / G] and increased the risk of individuals suffering of laryngeal squamous cell carcinoma, which may be the potential to predict the incidence of laryngeal cancer bio-markers.
