Objective:When rats explore different concentrations of lung ischemia-reperfusion protective effect of hormone erythropoietin, and further study the mechanism of rat lung ischemia-reperfusion.Methods:The 85 male rats were in good condition were randomly assigned into five groups(n= 17): pulmonary ischemia-reperfusion group(I / R), erythropoietin low concentration group(L), the concentration group(M), high concentration group(H), the control group(S). Before surgery 24 h, L, M, H groups were given by intraperitoneal injection of recombinant human erythropoietin(rh EPO) 1 000,3 000 and 5 000 U / kg, I / R group were injected sodium chloride injection; more than 4 hilar clamping vascular surgery group one hour after loose noninvasive hemostat 1.5 hours(S group does not clamp hilar vessels), observing the color of each set of lungs in rats, lung bleeding, blood gas analysis, rat lung wet weight and pathological changes in the mass ratio(W / D),production of paraffin, electron microscopy of rat lung after drying after HE staining,immunohistochemical staining was measured after rat lung monocyte chemoattractant protein-1(MCP)- 1 expression.Results:Comparison with the control group, and the remaining four groups of rats lung MCP-1 expression values and W / D values were increased(P <0.01), decreased arterialoxygen values in rats(P <0.01), and 4 groups with I / R group values changed significantly; among the three groups of erythropoietin pretreatment of rats lung M group compared with the numerical expression of MCP-1 L, H group reduced(P <0.05), W / D value than the L, H group(P <0.05), compared with arterial oxygen values L, H group increased(P <0.05).Conclusion:Different concentrations of the protective effect of erythropoietin in rat lung ischemia-reperfusion injury is not identical to, the optimal concentration is 3 000 U / kg,this protective mechanism of erythropoietin may inhibit excessive inflammatory MCP-1-mediated reaction.
