Objectives:To investigate the expression of the TGF-β1、CTGF and integrin β1 in vaginal anterior wall of the model rats with stress urinary incontinence.Methods:1.Thirty healthy female SD rats were randomly divided into three groups, group A rat as a control group breeded normally without any conduct;Conventional breeding experimental group B rats only a week after vaginal expansion method are adopted to simulate the childbirth, establish a model for stress urinary incontinence;Conventional breeding experimental group C rats by vaginal expansion plus a week after the bilateral ovaries removed the method to establish model for stress urinary incontinence.2.At postoperative four weeks to determine the maximum bladder capacity(MBC)and abdominal leak point pressure(ALPP), whether to establish detection model for stress urinary incontinence.3.Modeling successfully executed in rats after four weeks after vaginal anterior wall tissue, immunohistochemical staining method and real-time fluorescent quantitative RT-PCR technique to detect the expression of TGF-β1、CTGF and integrin β1.Results:1.A、B、C three groups of rats the maximum bladder capacity are(1.94 ±0.14)ml,(1.59±0.28) ml and(1.21±0.15)ml,A、B、C three groups of rats the abdominal leak point pressure are(28.86±5.01)mm Hg,(20.67±5.54) mm Hg and(14.36±5.07)mm Hg.Band C group is significantly lower in group A, and the comparison between groups have statistical significance(P<0.05).2.Three groups are visible in the vaginal wall tissue of TGF-β1 protein, the expression of CTGF protein and integrin β1 protein.TGF-β1, CTGF and integrin β1 protein is mainly expressed in cytoplasm, present a yellowish to brownish yellow brown cells is positive.TGF-β1 protein expression in three groups of vaginal wall immunohistochemical score(s) :(6.75±1.39),(5.13±1.81),(2.50±1.51), group C TGF-β1 protein immunohistochemical score(2.50±1.51), was lower than that in group A and B(6.75±1.39),(5.13±1.81), and was no significant difference between group A and group B(P > 0.05).CTGF protein expressed in three groups of vaginal wall of immunohistochemical score(s) :(7.75±1.49),(6.25±1.91),(5.50±0.93), and compare the difference between group A and group B with no statistical significance(P>0.05),significantly higher than that of group C, group A was statistically significant difference(P<0.05), and compare the differences between group B and group C has no statistical significance(P>0.05).integrin β1 protein expression in three groups of vaginal wall immunohistochemical score(s) :(6.13±1.36),(5.00±1.07),(3.63±0.74),and compare the difference between group A and group B with no statistical significance(P>0.05), group C was lower than that in group A, B two, compare the difference was statistically significant(P<0.05).3.RT-PCR determination results showed that TGF-β1 m RNA, CTGF m RNA and integrin β1 m RNA expression are all positive in the three groups.TGF-β1 m RNA in group A, B, C three vaginal wall tissue expression quantity respectively(1.04±0.35),(0.91±0.38) and(0.53±0.28), A group of TGF-β1 m RNA expression quantity relatively higher than that of group B, C, B group of TGF-β1 m RNA expression quantity is higher than that of group C(P<0.05), but there was no statistical difference between group A and group B more significance(P>0.05).CTGF m RNA in group A, B, C three vaginal wall tissue expression quantity respectively(1.05±0.42),(0.55±0.36) and(0.40 ±0.14), group B and group C CTGF m RNA expression of the mass were lower than group A, and the difference was statistically significant(P<0.05), but the comparison between group B and group C, there was no statistically significant difference(P>0.05).Integrin β1 m RNA in group A, B, C three vaginal wall tissue expression quantity respectively(1.03±0.28),(0.92±0.36) and(0.54±0.42), group C relative expression of integrin β1 m RNA is lower than in group A and group B, the difference was statistically significant(P<0.05), but the comparison between group A and group B, there was no statistically significant difference(P>0.05).4.Statistical methods: using SPSS 13.0 statistical software for statistical analysis,measurement data using mean±standard deviation, according to the comparison between the groups such as meet the normality and homogeneity of variance, and USES the single factor analysis of variance, the comparison between the LSD- t method;Is inferior to satisfy the normality and homogeneity of variance is used rank and inspection.Inspection level by α= 0.05, P < 0.05, the difference was statistically significant.Conclusion: Vaginal anterior wall in rats in SUI TGF-β1, CTGF and integrin β1 lower expression may be related to stress urinary incontinence and other pelvic floor support structure there is a close relationship of the occurrence and development of degenerative diseases.
