Objective To study the infuluence of SOCS hypomethylation on homeostasis of Th17/Treg in Henoch-Schonlein purpura(HSP),and the immune pathogenesis of HSPMethods 32 children with acute phrase HSP who were hospitalized from May to Decemeber 2014 in the Department of Cardiovascolar Kidney and Immunity Pediatric of Affiliated Hospital of Qingdao University were enrolled,28 healthy children from Child Health Division of the same hospital were served as controls.The levels of IL-6 in plasma were detected by ELISA.The proportion of CD4+CD+25、CD4+IL-17 A and mean fluorescence intensity(MFI)for phosphorylated-STAT3(p STAT3) protein in CD4+T cells was detected by flow cytometric analysis. Transcriptional levels of SOCS1,SOCS3 in CD4+T cells were assessed by real-time PCR.High-resolution melting(HRM) based on SYBR Green was used to evaluate methylation status of Cp G islands in SOCSl exon2.the potential bind sites for STAT3 in5’-untraslated region(5’-UTR) of SOCS3 in CD4+T ce Ils.Results 1.Compared with the control group,plasma IL-6 concentration, pSTAT3 and MFI for p STAT3 in CD4+T cells were remarkably increased in children with HSP(t=15.793,P<0.01;t=10.773,P<0.01;t=9.062,P<0.01);2.The levels of CD4+IL-17 A in CD4+T cells of the HSP children were significantly higher than the control group(t=7.585,P<0.05), but the levels of CD4+CD25+ in CD4+T cells of the HSP children were lower than the control group(t=4.582,P < 0.01).; 3.The m RNA levels of SOCSl and SOCS3 in CD4+T cells increased significantly during acute phase of HSP(t=8.436,P<0.01;t=8.153,P<0.01);the SOCS1,SOCS3 m RNA expression rates of peripheral blood mononuelear cell and the proportion of Th17 and Treg cells had negative correlation, the expression of p STAT3 in CD4+T cells and the proportion of Th17 and Treg cells had positive correlation; 4.Cp G isislnds in SOCSl exon2 and the potential binding site for STAT3 in SOCS3 5’-UTR were hypomethylated in acute HSP,While those in control group were fully demethylated.Conclusions Relative insufficiency of SOCS1 and SOCS3 expression caused by hypomethylation may be one contributing factor for the imbalance of Th17/Treg in HSP.
