BACKGROUND: Studys have found that the liver cells can synthesize insulin after giving into PDX1 gene. Anti-CD20 monoclonal antibody can inhibit the cell immune reaction of liver-derived insulinproducting, but the mechanism is unclear.OBJECTIVE: To observe the inliuence of liver cells and liver PDX1 expression condition in Nonobese diabetic mice which interfered by Adenovirus vector-mediated murine interleukin and Anti CD20 monoclonal antibody.METHOD: 40 NOD mice,3-5 weeks, female, were randomly divided into(1) group Anti-CD20,(2) group Anti- CD20 + IL- 10,(3) group IL- 10,(4) group physiological saline(Normal control, NC). Injected Anti- CD20, Anti- CD20 + IL- 10, IL- 10 and physiological saline on day 1, day 8, day15 and day 21 via tail vein. Monitored blood glucose weekly, detected serum Insulin, IL-10 and CD20 level after 12 weeks.Observed the pathological HE staining of the liver cells. Monitored the expression of IL – 10, CD20, Insulin and PDX1 of liver cells in immunohistochemical method.RESULTS AND CONCLUSION : Association of Anti- CD20 and IL- 10 can control the blood glucose of NOD mice effectively,can increase the serum insulin, IL- 10 and CD20 expression level, and can promote the expression of insulin, CD20, IL – 10 and PDX1 in liver cells. But the intervention had no obvious effection on inflammation of liver cells.
