Objeetive:1.To establish the model of cyclophosphamide induced osteoporosis rats.2.To observation the effects of zuoguiwan on Bone turnover markers, bone mineral and bone microstructure in Cyclophosphamide induced osteoporosis rats. To investigate the mechanism of zuoguiwan in the treatment of osteoporosis.For the clinical use of zuoguiwan Treating Osteoporosis and provide experimental theoretic basis. Methods:40 SD female rats of SPF aged 3 months, weight(200 士 20)g,after adaptive feeding a week, in accordance with the random number table method, according to the weight, were randomly divided into blank control group(n=10) and Osteoporosis model group(n=30). The rats in the Osteoporosis model group were intrapentonal injection cyclophosphamide(8mg/kg), control group were intrapenitoneal injection the same volume of normal saline,once a day,for 15 days. Then the rats were in bone density testing confirmed the model group rats osteoporosis model was successfully built. In accordance with the random number table method, according to the weight, the Osteoporosis model group is divided into cyclophosphamide group(n=10), zuoguiwan group(n=10), Calcium group(n=10). The rats in the control group with 2ml normal saline by gavage, once a day,for 4 weeks. The rats in cyclophosphamide group with 8mg/kg does of cyclophosphamide by gavage, once a day,for 4 weeks. The rats in the zuoguiwan group with 18mg/kg does of zuoguiwan by gavage, once a day,for 4 weeks. The rats in the Calcium group with 3mg/kg does of Calcium gluconate by gavage, once a day,for 4 weeks. weighed once every 3 days, adjust the dose based on body weight. After 4 weeks of treatment, detection of markers of bone turnover, bone density, trabecular,bone microstructure and other indicators. Results: 1. To establish the model of cyclophosphamide induced osteoporosis rats:Cyclophosphamide injection after 15 days, different dose of cyclophosphamide group compared with control group, the whole body bone mineral density,lumbar bone mineral density spine, femoral bone mineral density tended to decrease. Middle and high doses of cyclophosphamide group compared with the control group, the whole body bone mineral density,lumbar bone mineral density spine, femoral bone mineral density were statistically significant(P < 0.05); Different dose of cyclophosph-amide group compared with control group, bone volume fraction, trabecular number, trabecular thickness reduced, the difference was statistically significant(P < 0.05). Trabecular separation, structure model index rise, the difference was statistically significant(P < 0.05); Different dose of cyclophosphamide group compared with control group, serum calcium, phosphorus decreased, the difference was statistically significant(P < 0.05). Alkaline phosphatase, osteocalcin increased, the difference was statistically significant(P < 0.05). 2. The effects of zuoguiwan on cyclophosphamide induced osteoporosis rats:(1) Weight change:cyclophosphamide group compared with control group,weight is significantly reduced, the difference was statistically significant(P < 0.05).zuoguiwan group and Calcium group compared with control group,weight is significantly increased, the difference was statistically significant(P ﹥0.05).(2) Organ index change: the rats liver index, spleen index were not significantly difference. zuoguiwan group and Calcium group compared with control group,thymus index was significantly increased, the difference was statistically significant(P < 0.05).(3) Detection of serum trace elements: each group rats had no significant changes in serum magnesium. control group,zuoguiwan group and Calcium group compared with cyclophosphamide group, Serum calcium, phosphorus, zinc increased, the difference was statistically significant(P < 0.05). The blank control group, zuoguiwan group and calcium group between three groups of serum calcium, phosphorus, zinc, no obvious difference.(4) Bone turnover markers: cyclophosphamide group compared with control group,zuoguiwan group and Calcium group, Osteocalcin and alkaline phosphatasewere increased, the difference was statistically significant(P < 0.05). The blank controlgroup, zuoguiwan group and calcium group between three groups of Osteocalcin, no obvious difference;(5) Bone mineral density change: between the four groups before and after treatment of rats bone density, bone mineral density of the lumbar spine, femoral bonemineral density had significant difference(P < 0.05). Compared with cyclophosp-hamide group, zuoguiwan group, calcium group rats after treatment of bonedensity, bone mineral density of the lumbar spine and femoral bone mineral density obviously improved, the difference was statistically significant(P < 0.05).Compared with control group, zuoguiwan group and Calcium group bone density,bone mineral density of the lumbar spine and femoral bone mineral density no obvious difference.(6) Determination of bone microstructure parameters: Cyclophosphamide group bone volume fraction, trabecular number, trabecular thickness were lower than the other three groups, the difference was statistically significant(P < 0.05).Cyclophosphamide group trabecular separation and structure model index higher than the other three groups, the difference was statistically significant(P < 0.05).(7) Trabecular change: The rats in zuoguiwan group and control group trabecular arranged closely, regular,normal bone marrow cavity. The bone mineral density of cyclophosphamidegroup rats decreased significantly, decreased trabecular bone volume, trabecular bone thinning, thinning, arranged disorder, trabecular bone transformation from plate to rod. Conclusion: 1. Cyclophosphamide can make the bone mineral density of rats decreased, bone microstructure damaged, Serum calcium and phosphorus decreased, markers ofbone formation Elevated. Effect of cyclophosphamide on human bone tissue is simulated. Description model of cyclophosphamide induced osteoporosis rats model success. 2. Zuoguiwan can affect the calcium and phosphorus metabolism in rats, inhibit bone resorption, promote bone formation, increase bone mass, increase the bone density,bone trabecula internal shape optimization, improve bone microstructure, increased bone strength. The said findings indicate that zuoguiwan is an effective way to prevent osteoporosis, worthy of popularized application.
