Objective:This experiment aims to observe apoptosis change of nerve cells which treatedwith different dose of angelica sinensis on rat models.Methods:1.Group of Experiments:Rats were randomly divided into 6 groups(n=10):control group,sham-operated group,model group,angelica sinensis 2.8g/kg treated group,angelica sinensis 5.4g/kg treated group,angelica sinensis 10.8g/kg treated group.2.Rat model of cerebral hemorrhage establishment and drug intervention:Type Ⅳ collagenase was injected into rat caudate nucleus to obtain ra t model of cerebral hemorrhage.control group free from any treatment,sham-operated group just treated with craniotomy,Ⅳ collagenase just treated model control group, model control group and sham-operated group just treated with distilled water every day.Different dose of angelica sinensis treated group(2.8g/kg、5.4g/kg and 10.8g/kg)were gavaged every day.3.Score of nerve dysfunction:neurobehavioral score was evaluated with Bederson grades methods 4.Samples test:RT-PCR was used to measure the level of Bax m RN A,Bcl-2 m RNA and Caspase 3 m RNA.Neural cell apoptosis related proteins Bax、Bcl-2 and Caspase-3 were measured with immunohistochemical method.5.SPSS 17.0 software was used for all analysis.The difference of data between different groups compared with one-way ANOVA analysis. P<0.05 was considered to be statistically significant.The values were expressed as mean±sd.Results:1.Immunohistochemical results show that the Bax and Caspase-3 decreased and Bcl-2 increased in a dose dependent manner in intracerebral hemorrhage rats which were treated with different doses of angelica sinensis.2.RT-PCR results show that the Bax m RNA and Caspase-3 m RNA decreased and Bcl-2 m RNA increased in a dose dependent manner in intracerebral hemorrhage rats which were treated with different doses of angelica sinensis.Conclusion:Angelica sinensis could inhibits nerve cells apoptosis on cerebral hemorrhage rats. Expression of apoptosis related protein included Bcl-2,Bax and Caspase–3 were involved in apoptosis mechanism.
