ObjectivesThis article aims to detecte the expression of Ezrin and P21 in normal breast tissues 、 mammary induct carcinoma and invasive ductal carcinoma of breast,respectively,Explore whether the levels of two proteins are different in those tissues or not, and confirm the relationship between the two proteins and the other clinical characteristics and pathological markers of invasive ductal carcinoma of breast. Further to study the correlation of the expression levels of two proteins in different breast cancers and their metastasis and recurrence and prognosis.Materials and methodsRandomly selected 94 pathological tissues that have been fixed by paraffin in the first affiliated hospital of Nan Chang University from January 2012 to January 2014.There are 22 normal breast tissues,32 mammary induct carcinoma and 40 invasive carcinoma of breast,respectively.We have detailed clinical and pathological data of all tissues and the follow-up data. The patients are not treated by chemoradiotherapy and hormonal therapy before surgery, and these tissues are incised on the first operation.The table 1 shows the data. Test the expression levels of Ezrin and P21 in tissues by Envision, and analyze the data by combining clinical and pathological parameters.Results1.Ezrin is positive dyed brown yellow and sepia in invasive breast cancer, which is focus on the cell plasma and pervasive.The expression rate of Ezrin is 72.5%(29/40) in invasive breast cancer. The positve rate of Ezrin is 40.7%(13/32) in intraductal carcinoma,which is pervasive focus on the cytoplasm and dyed brown yellow and sepia. However, Ezrin is dyed pale yellow in normal breast cell, which is focus on the membranes of ductal epithelium, the positve rate of Ezrin in plasma is 13.6%(3/22).The expression of Ezrin is most in invasive breast cancer,secondly in mammary induct carcinoma and finally in normal breast tissues,there is statistical significance comparing between any two.2.P21 is positive dyed brown yellow in cell nucleus or in cell nucleus and plasma,but P21 is negative dyed yellow only in plasma.The positive rate of P21 is 67.5% in invasive breast cancer(27/40),34.4% in mammary induct carcinoma(11/32),13.6% in normal breast tissues(3/22).P21 becomes lower and lower in invasive breast cancer 、 mammary induct carcinoma and normal breast tissues,there is significantly difference comparing between them.3.The expression rate of Ezrin is higher in stage III-IV(11/12,91.7%) than stage I-II(18/28,64.3%) in invasive breast cancer,and the expression rate of Ezrin is higher in the lymphatic metastasis greater than or equal to 4(100%) than in the lymph node metastasis less than 4(46.4%),there is statistical significance(P<0.01). However, the expression rate of P21 is lower in the stage III-IV(4/12,33.3%) than in the stage I-II(23/28,82.1%) in the invasive breast cancer, it shows significantly difference(P<0.01);the expression rate of P21 is higher in the lymph node matestasis greater than or equal to 4 than in the lymph node metastasis less than 4,but there is no statistical significance(P<0.01).4.Ezrin and P21 are highly expressed in the invasive breast cancer,especially,the expression of Ezrin is higher in the cases that have lymphatic metastasis, and the correlation between the expression of Ezrin and number of lymphatic metastasis is positive.5.The expression of Ezrin in the breast cancer is positive correlation to histological grade 、 number of lymphatic metastasis and age, but is negative correlation to PR.The expression of P21 in the breast cancer is negative correlation to TNM stage、histological grade and the expression of Her-2,but is positive correlation to PR.The expresion of P21 and Ezrin in the breast cancer is no correlation to menopausal status、tumor size、ER status. There’s no correlation between Ezrin and P21 in the breast cancer.Conclusions:1.Ezrin has different subcellular locations in the benign and malignant breast diseases, the expression of Ezrin plays an important role on the judgement of the breast diseases’ nature.P21 is highly expressed in the breast cancer, locates in the cell nucleus,occasionally in the cell plasma.Ezrin beomes lower and lower in the invasive breast cancer、mammary induct carcinoma and normal breast tissues, there’s statistical difference,so does the P21.2.The expression of Ezrin and P21 is higher in invasive ductal breast cancer than mammary induct carcinoma,it shows that they are congenerous to promote malignancy and metastasis in the tumerigenesis and development of invasive ductal breast cancer.3.The expression of Ezrin in the breast cancer is positive correlation to histological grade 、number of lymph node metastasis,but is negative correlation to PR;The expression of P21 in the breast cancer is negative correlation to TNM stage、histological grade and the expression of Her-2,but is positive correlation to PR.It indicates that they are related to the development 、metastasis and prognosis of breast cancer,joint detection of them can predict the prognosis of breast cancer,and provide reference value in the targeting therapy of breast cancer.
