ObjectivesThis study try to design two kinds of surgical methods in producing corneal endothelial graft of rabbit by the diversity cutting modes of Femtosecond laser,and evaluate the feasibility and safety of these two surgical modes according to the results of survival status of graft in autologous cornea and Laboratory tests on the vitro tissue after surgery.Method8 Health New Zealand white rabbits without eye disease,male and female,weight ranging from 2kg to 2.5kg,randomly divided into 2 groups after examed by anterior segment slit lamp microscope photography,Corvis ST,Anterior segment OCT and Non-contact corneal endothelial cell analyzer, every group have 4 rabbits,choose the right eye for the surgery. A group is Mode 1(Full Lamellar Cut and Posterior Side Cut)surgery group,B group is Mode 2(Full Lamellar Cut and Anterior Side Cut)surgery group. Set parameter valuesa of each surgical mode in a FS150 Femtosecond laser apparatus to produce corneal endothelial graft. Using tobramycin and dexamethasone eye drops for 2 weeks after surgery. Respectively do the anterior segment slit lamp microscope photography,Anterior segment OCT and Non-contact corneal endothelial cell analyzer test since 1 week,1 month and 3 month after surgery. Rabbits were killed by ear vein of air embolism at 3 month after surgery and HE staining on the corneal tissue, TUNEL test to detect Corneal endothelial cell apoptosis detection.Results1. There is no statistically significant difference between CCT measure by Corvis ST and Anterior segment OCT(p>0.05). 2. Result of anterior segment slit lamp microscope photography exam on corneal surgery area at 1 week,1 month and 3 month after surgery is transparent andwithout edema both in group A and B,no corneal infection and other complications appear. 3. Result of Anterior segment OCT exam on the corneal endothelial graft at 1 week,1 month and 3 month after surgery is normal both in group A and B,there is no statistically significant difference in thickness change of the corneal endothelial graft compared with the setpoint before surgery(p>0.05). 4. There is no statistically significant difference between ECD,CV and HG at 1 week,1 month and 3 month after surgery compared with the preoperative neither in group A nor B(p>0.05). 5. Both in group A and B, corneal endothelial cells holding a normal morphology in HE staining. 6. There is no corneal endothelial cell apoptosis detection neither in group A nor B from TUNEL test.ConclusionThere is a good consistency in CCT measure result between Corvis ST and Anterior segment OCT, so we can use the CCT result of Corvis ST to set depth parameter valuesa in Femtosecond laser surgery. Corneal endothelial graft and corneal endothelial cells are in good condition during observation period after surgery in both two surgical modes,these two surgical modes has satisfactory surgical results and considerable prospects.
