China has a vast territory with favorable natural conditions. There is an abundance of traditional medicine resources in China with the longest history of herb cultivation all over the world. Since Chinese people accumulated rich experience in the fight against disease process through constant practice and study for thousands of years, part of the Chinese herbal medicine has been proven to have an exact curative effect. With the development of productive forces and social progress, people are increasingly looking forward to develop much more safe and effective new drugs from natural sources. So Chinese herbal medicine researchers must illustrate the active ingredients of Chinese herbs to establish the basic research for improving quality and efficacy.Vitex rotundifolia and Cassia mimosoides are commonly used in traditional Chinese medicine in China, but their material basis of drug effects is still unclear. This dissertation consists of two chapters, in which their chemical constituents and biological activity were studied. Total 47 compounds were isolated and identified by the chromatography, spectrum methods, physical and chemical properties, among which 13 compounds were new. Some of them showed moderate cytotoxic activities.Twenty diterpenoids were isolated from Vitex rotundifolia, including 11 halimanoids, 6 labdanoids, 2 rearranged halimanoids and 1 clerodanoid, of which 11 were new compounds(including 2 rearranged halimanoids, 7 halimanoids, 1 labdanoid and 1 clerodanoid). The new compounds were named as viterofolin A~K. The cytotoxic activity test(MTT assay) was conducted for all the isolated compounds. Compound 2 showed moderate cytotoxic activities against A375(BRAFV600E mutation melanoma cells) and HCT-116(no BRAFV600 E mutation colorectal cells), with the IC50 of 69.84 and 49.01 μM respectively. Compound 19 also showed moderate cytotoxic activity against A375 with an IC50 of 45.19 μM.27 compounds were isolated and identified from Cassia mimosoides, including 3 guanidine alkaloids, 6 norsesquiterpenes and its glycosides, 5 flavones and its glycosides, 5 phenolic acid glycosides, 5 phenypropanoid glycosides, 2 lignan glycosides and 1 iridoid glycoside. Compound 21 and 27 were new compounds. In addition, the guanidine alkaloids were rarely found in the terrestrial plant. The isolation of 3 guanidine alkaloids in Cassia plant layed foundation of looking for more guanidine alkaloids.
