Objective:To study the expression of the Matrix Metalloproteinases 2(MMP-2) and Matrix Metalloproteinas 9(MMP-9) in human hypertrophic scarfibroblasts which were applied by thymosin beta4,in order to know the possible mechanism of thymosin beta4 on the morbidity of pathological scar. Methods:Select the patients in department of burns and department of plastic surgery in our hospital. The criteria for inclusion was:(1) burns happen six months before;(2) Scar appeared more severe contracture;(3) Other drugs and other treatments are not not used;(4) There are surgical indications and surgery was performed in our hospital;(5) Patients and family members voluntarily signed relevant informed consent. The experiments take the Hypertrophic scar fibroblasts(HSFB) after 3-6 generations. The cells were divide into five groups, the control group: no thymosin beta4 added, the experimental group: Add 0.05μ g/m L,0.1μ g/m L, 1.0μ g/m L,5.0μ g/m L of thymosin beta 4 intervention for 24 hours, respectively. We detected the expression of MMP-2 m RNA and MMP-9 m RNA by RT-PCR, and the expression of MMP-2 and MMP-9 protein by Western blot. Results:(1) Compared with control group, Thymosin beta 4 could increase the expression of MMP-2 m RNA and MMP-9 m RNA in each experimental group, the effect of increasement of Thymosin beta 4 with 5.0 μ g/m L was most significant. The difference was statistically significant(p<0.05).(2) Compared with control group, Thymosin beta 4 could increase the expression of MMP-2 and MMP-9 protein in each experimental group, the effect of increasement of Thymosin beta 4 with 5.0μg/m L was most significant. The difference was statistically significant(p<0.05). Conclusion:Thymosin beta 4 could increase the expression of MMP-2 and MMP-9 in HSFB. And thymosin beta4 may degradation and reduction of the extracellular matrix.
