Objective: To detect the expression of Wnt1, Wnt5 a and Survivin in pancreatic ductal adenocarcinoma(PDAC) and normal pancreatic tissue adjacent to carcinoma, study its relationship with the clinicopathologic parameters of PDAC and explore its clinical significance.Methods: 43 cases of PDAC tissues and corresponding normal pancreatic tissues adjacent to the carcinoma, which Formalin- fixed and paraffine-embedded were collected from the department of pathology of the First Affiliated Hospital of Nanchang University from January 2008 to December 2013. The expression of Wnt1, Wnt5 a and Survivin were detected by streptavidin-perosidase(SP) immunohistochemical method. The statistic analysis of experimental data was performed by SPSS 19.0 software.Results: 1. Wnt1 was mainly expressed in cytoplasm. The expression of Wnt1 in PDAC tissues and normal pancreatic tissues adjacent to carcinoma were 72.09%(32/43) and 34.88%(15/43), respectively. The expression of Wnt1 in PDAC group was significantly higer than that in normal pancreatic group(P<0.05). The expression of Wnt1 in PDAC group was significantly correlated to histological grades, nerve invasion, lymphatic metastasis and TNM stage(P<0.05), but was not correlated to age, sexuality, tumor size and tumor location(P>0.05). 2. Wnt5 a was mainly expressed in cytoplasm. The expression of Wnt5 a in PDAC tissues and normal pancreatic tissues adjacent to carcinoma were 69.77%(30/43) and 27.91%(12/43), respectively. The expression of Wnt5 a in PDAC group was significantly higer than that in normal pancreatic group(P < 0.05). The expression of Wnt5 a in PDAC group was significantly correlated to nerve invasion, lymphatic metastasis and TNM stage(P<0.05), but was not correlated to age, sexuality, tumor size, tumor location and histological grades(P>0.05).3. Survivin was mainly expressed in cytoplasm. The expression of Survivin in PDAC tissues and normal pancreatic tissues adjacent to carcinoma were 81.39%(35/43) and 11.63%(5/43), respectively. The expression of Survivin in PDAC group was significantly higer than that in normal pancreatic group(P < 0.05). The expression of Survivin in PDAC group was significantly correlated to histological grades, nerve invasion and lymphatic metastasis(P<0.05), but was not correlated to age, sexuality, tumor size, tumor location and TNM stage(P>0.05). 4. The Spearman correlation analysis showed a positive correlation between the expression of Wnt1 and Survivin in PDAC(r=0.542, P<0.05). But there have no correlation between the expression of Wnt5 a and Wnt1(r=0.194, P>0.05), and also no correlation between Wnt5 a and Survivin(r=0.186, P>0.05).Conclusions: The abnormal expression of Wnt1, Wnt5 a and Survivin were closely associated with the progress of pancreatic cancer. Survivin was probably one of the Wnt1 mediated Wnt signaling pathway downstream target genes. Combined detection of Wnt1, Wnt5 a and Survivin protein could help to evaluate the pro gnosis of pancreatic cancer and provide theoretical basis for treatment.
