Purpose: To test the expression of CKl7,and P16 in CIN and cervical squamous cell cancer,at the same time to discuss the relationship between them.Methods:.Select 20 cases of normal cervical tissues or the nonspecific inflammation tissues, CIN group 53 cases(including 15 cases,of CINI,17 cases of CINⅡ, 21 cases,of CIN Ⅲ), 42 cases of early cervical squamous carcinoma group. collecting the general data, such as the age, clinical stage, pathological grading.Using the S- P staining to detect the expression of CK17 and P16,and put the normal cervical tissues as control.Statistic and analysis the expression between CK17 and P16 in different tissues of the cervical lesions, and to explore the relationships.Results: 1.The changes of CK17 in different cervical pathological tissueIn this experiment, the expression of CKl7 is located in cytoplasm, mainiy expressed in cervical reserve cell, it was not expressed in normal cervix epithelia, the expression rate is lower than the diseased tissue, its early in normal cervical tissue and CIN and cervical squamous cell carcinoma of the positive expression rate is 10%, 49.1% and 73.8%.The difference among the three groups was statistically significant(c2= 22.280, P < 0.05).In each group of the CIN cases the positive rate of CK17 was 26.7%、41.2%、71.4%, and the difference was statistically significant(c2= 7.637, P <0.05). Group comparison, CINIII group was higher than CINI group, the difference was significant, CINII group have not statistically significant.with CINⅡand CINⅢ.In early cervical squamous cell carcinoma CK17 have no relationship between the degree of cervical clinical expression and tissue differentiation in cervical cancer. In the three groups of cervical tissue,the expression rates of CK17 were 10%, 49.1% and 73.8%,positive rate was correlated with the degree of cervical lesions(r = 0.931 P <0.05) 2.The changes of P16 in different cervical pathological tissueIn this experiment, the expression of P16 is located in cytoplasm or nucleus. Its in normal cervical tissue and CIN and early cervical squamous cell carcinoma of the positive expression rate were 5%, 75.5% and 95.2%.respectively, The difference among the three groups was statistically significant(c2=54.176, P < 0.05)..In each group of the CIN cases the positive rate of CK17 was 50%、76.5%、90.91%, and the difference was statistically significant(c2=8.910, P <0.05). Group comparison, CINIII group was higher than CINI group, the difference was significant, CINII group have not statistically significant.with CINⅡand CINⅢ.In early cervical squamous cell carcinoma CK17 have no relationship between the degree of cervical clinical expression and tissue differentiation in cervical cancer(P>0.05)In the three groups of cervical tissue,the expression rates of P16 were 5%, 75.5% and 95.2%,positive rate was correlated with the degree of cervical lesions(r = 0.888 P <0.05). 3.The relationship between CK17 and P16 expression in cervical tissue lesionsIn this study, 125 cases of cervical lesions slice organization, there were 57 cases of CK17-positive expression, ⅠA of positive were 9 cases P16 of positive were 80 cases, including 41 cases of lesions were positive for CK17 and P16 expression can be derived by analyzing the correlation between them the correlation coefficient r = 0.743 P <0.01 Description CK17 expression was significantly correlated with P16 in cervical lesions tissues.Conclusion1.CK17 express only in the normal reserve cells. in CIN and early cervical squamous cell carcinoma, the expression rate was elevated with the increasing lesion grade of the intraepithelial.it was significantly expressed in cervical squamous cell carcinoma,major located in parts of the tumor thrombus and the infiltration of frontier area, we can conclusion that it can be used as a marker of cancer stem cells.2.The P16-positive can well differentiate normal cervical hyperplasia, CIN and early cervical squamous cell cancer, especially for distinguishing benign from malignant cervical lesions and to determine the clinical stage of disease, it can be used as a good indicator for the early diagnosis of cervical lesions.3.Combined the CK17 and P16 can play a complementary role, CK17 can identify cervical IM and CIN, P16 can identify benign and malignant cervical trends, combined detection can be a good predictor for the development of benign and malignant cervical lesions trends.
