Objective:To investigate the levels of anti- BRAF antibody expressing in serum of the RA patients and its clinical significance.Methods:We examined the level of anti-BRAF antibodies in 95 patients with RA,88 patients with other rheumatic diseases and 48 healthy controls by enzyme-linked immunosorbent assay(ELISA). The clinical data of each group were collected simultaneously.Then we can investigate the relationship between the levels of anti- BRAF antibody and the clinical symptoms, results of laboratory and the related imaging changes of the RA group.Results: ① The level of anti- BRAF antibody in serum of RA was significantly higher than other rheumatic diseases groups and healthy controls(Z=-5.213,P<0.01;Z=-4.335,P<0.01). ② The anti-BRAF antibody of the ROC curve area(AUC) are 0.723±0.039(95%CI:0.646-0.800),the best diagnostic boundaries of serum anti- BRAF antibody was OD>0.4, the sensitivity of prediction was 58.947%, and the specificity was 92.6%.③ The positive rate of anti-BRAF antibody was 58.95% in RA patients, which was significantly higher than that of ankylosing spondylitis(12.5%,P<0.01) and systemic lupus erythematosus(15%,P<0.01), was significantly higher than that of norma control group( 8.33%, P<0.01). ④ The positive rate of anti-BRAF antibody in Rheumatoid Factor negative group was significantly higher than that of the positive group in RA.(χ2=7.891,P<0.01). ⑤ The positiv rate of anti-BRAF antibody in joint swelling group was significantly higher than that of the without joint swelling group(χ2=5.508,P<0.05)in RA. It showed a negative correlation between the level of anti-BRAF antibody and patient’s disease duration and the level of Hemoglobin in RA. ⑥ The X-ray of hands shows that there is no significant differences in anti-BRAF antibody positive rate between normal group and bone destruction group(χ2=1.091,P>0.05). ⑦ The computed tomography of chest shows that there is no significant difference in anti-BRAF antibody level between normal group and pulmonary fibrosis group(χ2=0.062,P>0.05).Conclusion:① Anti-BRAF Ab has shown a high positive rate in the patients with RA. It can be used for the diagnosis of RA as a new autoantibody. ②The anti-BRAF Ab has shown a high positive sequence in the RA paitents who are found in the early disease and those with negative serum test. ③The level of Anti-BRAF Ab has a positive correlation with disease activity. ④The level of Anti-BRAF Ab has no significant correlation with joint bone destruction and lung affected.
