Objective: To explore the role and mechanism of helper T cells 17(Th17) in paraquat poisoning.Methods: To observe the changes in Th17 cells in paraquat poisoning in mice, we inject paraquat 35mg/kg in mice abdominal cavity to build the poisoning model in this experimental study, five days later using flow cytometry(FCM) to observe the changes of Th17 cells, retinoic acid orphan receptor gamma t(RORĪ³t)in paraquat poisoning mice; In order to further confirm the role of Th17 cells in paraquat poisoning mice, On the-1 days(one day before building the poisoning model) inject anti IL-17 antibody in mice abdominal cavity to neutralize IL-17, 0 days poisoning mice model was established, 5 days to observe the changes of Th17 cells,cytokine, lung tissue pathology, and the survival rate, to explore the role and mechanism of Th17 cells in the development of paraquat poisoning disease; Finally we used flow cytometry to observe Th17 cells in blood of paraquat poisoning patient.Results: By using flow cytometry, polymerase chain reaction(PCR), protein imprinting(Western Blotting)method to shown Th17 cells, transcription factor ROR gamma gene and protein expression increasing significantly on five days after building the poisoning modle. Utilize IL-17 antibodies can neutralize Th17 cells and increase survival rate, reduce the lung tissue injury in paraquat poisoning mice. Using flow cytometry we found that Th17 cells increas significantlyin patients with paraquat poisoning blood than the normal group.Conclusion:Th17 cells play a role of proinflammatory and aggravate the damage in paraquat poisoning.
