Object: Alzheimer’s disease(AD) is a degenerative neuropathy characterized by mental retardation, but its etiology and its mechanism are not clear. Cholinergic hypothesis which has been a hot research of scholars domesticly and abroad, and guide people to AD knowledge and therapeutic intervention. Acetylcholine(ACh) as one of the most important neurotransmitters in the central nervous system is widespread in the brain, and its main function is to maintain a clear sense, plays an important role in learning and memory. By many test results, more and more scholars believe that the central cholinergic system damage raised from all kinds of causes may be the common basic mechanism of dementia. Our study was to investigate changes in protein expression in brain tissue in Alzheimer’s disease model rats, and changes in neuron specific enolase(NSE) expression in the serum of patients with vascular dementia.Method: 20 Wistar male rats selected as sample in the experiment, were randomly divided into normal group and AD model group. Cutted off bilateral hippocampal and prepared animal model of AD. Peptide fingerprinting is that biological mass spectrometry analysis and indentification of specific protein spots were conducted by NCBI. Hippocampal proteins in different experimental groups were detected. By comparison of protein changes(conditions: AV Ratio> 1.3 or <1.3), the spots(P <0.05) were recognized as meaningful spots. The results were further identified by Swiss-Port protein database. Statistical analysis was performed by the comparison of NSE expression in serum of normal peoples and in that of patients with VD.Result: A significantly increased expression of proteins in the hippocampus of AD model group were found by peptide mass fingerprint spectra, identified as NSE. NSE expression in serum of patients with VD is higher than that of normal.Conclusion: NSE expression were changed in the course of AD. NSE was the important enzyme in AD disease and also changed in the serum of patients with VD.
