Objective: To identify the different expressing protein spots among melanosis coli(MC), colon cancer and normal tissues by two-dimensional gel electrophoresis(2-DE), and to explore the occurrence and development of MC and its correlation with colon cancer.Methods: 1. Tissue samples were selected from our hospital endoscopy room from January 2013 to June 2014, including 7 cases of MC group, 6 cases of colon cancer group and 6 cases of normal control group. All these samples were identified by pathological diagnosis; 2. After making colon tissue samples, the protein concentration of the supernatant was measured by the Bradford method, and then established twodimensional gel patterns of MC, colon cancer and normal colon tissue by twodimensional gel electrophoresis(2-DE) technique that first step was the immobilized pH gradient isoelectric focusing(IEF) and the second was sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE); 3. After coomassie-staining, the images of gels were acquired by the scanner and analyzed using imagemaster 7.0 software to find different expressing protein spots between melanosis coli and colon cancer, normal tissue.Results:1. Gel images that obtained from the samples showed good image reproducibility. The protein spots were clearly separated, each group had a good repeatability in the location of protein spots on the image and the image could be performed; 2. Average protein spots of colon cancer, melanosiscoli and normal tissues were respectively 2150±240, 1970±138, 2028±176. Compared with gel patterns, we found 14 points in difference with the confidence level of 95%. Compaired MC with normal tissue, there were six points in difference, including four protein spots were upregulated in normal tissues, two points upregulated in MC. And there was one protein that was only expressing in MC(P<0.05); There were 8 points of difference between MC and colon cancer, including one protein spot was upregulated in MC, seven points upregulated in colon cancer. And there were two proteins that were only expressed in colon cancer(P<0.05).Conclusion: We analyze the proteomics of MC and colon cancer by 2-DE and comparative analysis of gel images. There are differentially expressed proteins between MC and normal colon tissue, colon cancer tissue after two-dimensional gel electrophoresis and image analysis, which may reveal the biological behavior including occurrence and development of MC and colon cancer, and may provide new ideas or clues for the study of MC.
