Objective To investigate the degree of vascular restenosis with time in rabbits with graft vein and the therapeutic effect of argatroban on the vascular restenosis, the expression of PCNA in the vein bridge, eliude the mechanisms of argatroban for treatment the graft vein restenosis and curative effect.Methods Three months old, twenty rabbits(weight 2.5~3.0Kg), all of males, were were randomly divided into A, B, C and D groups( 5 rabbits each group) by random number table.All animals were subjected to ipsilateral common carotid artery-vein graft surgery models with sodium pentobarbital anesthesia, and the rabbits were injected the saline and agratroban0.667ml/Kg/d in vein from the second day after surgery in A and B groups, C and D groups,respectively. The rabbits in A and C groups, B and D groups were sacrificed by air injection on the fourteenth day and the twenty-eight day after surgery, the graft vein was excised and the vien bridge separated and rinsed with physiological saline, several microtome sections were collected from each tissue for morphological observation, the thickness of intima and media were calculated from the sections stained with HE by IPP software analysis and the PCNA in media were assayed in vascular intima by immunohistochemical procedure, assessed the postive index(PI) value by IPP softwave. Statistical analyses were performed using SPSS version 13.0 to compare the the thickness of neointima and media, and the differential expression of PCNA in vein bridge media. Differences between groups were considered significant when P≤ 0.05.Results The morphological comparison of cartilage tissue:In A and B groups, the thickness of intima and media were significant incrassation, it were significantly higher in A and B groups than C and D groups(p<0.001, p<0.05; respectively). it were significantly higher in B group than A group(p<0.05). The the thickness of intima and media in C group were increased than D group, butit were insignificantly higher in C group than D group(p>0.05);The PI of PCNA expression in vein bridge media were significantly higher in A group and B groups than C and D groups(p<0.01, p<0.05, respectively). The PI of PCNA expression in vein bridge media were significantly higher in B group than A group(p<0.05);The PI value of PCNA expression in vein bridge media were increased in C group than D groups, butit were insignificantly higher in C group than D group(p>0.05).The the thickness of intima and media, PI of PCNA expression in vein bridge media were significantly increased in A group than C group(p<0.01 or p<0.05,respectively). The the thickness of intima and media, PI of PCNA expression in vein bridge media were significantly increased in B group than D group(p<0.01 or p<0.05, respectively).Conclusions 1. Tansplant the common carotid vein to common carotid artery could success in making the stenosis of transplanted vessels model. With the time increasing, the morbidity of stenosis of transplanted vessels is aggravating. 2. Argatroban could inhibit the development of the stenosis of transplanted veins in early phase. Adminstrate the argatroban over a certain period of time after prolonged treatment does not further improve curative effect. 3. One of the mechanisms of argatroban prevent the development of the stenosis of transplanted veins maybe down-regulated the expression of PCNA in the veins bridge.
