Objective:Establish the model of atherosclerosis in rats, observe the effect of Lycium barbarum polysaccharides Component Ⅲ(LBP-Ⅲ)on the rat model of atherosclerosis and investigate its mechanism in order to provide experimental basis for the prevention and treatment of drug on AS.Methods:60 healthy rats(male SD rats aged 4~5 months at the clean level) were randomly divided into the following 5 groups(n=12): normal control group; AS model group; the lovastatin group(lovastatin 6mg/kg/day,gavage); the low dose LBP-Ⅲgroup(0.5934g/kg/ day, gavage); the high dose LBP-Ⅲgroup(1.875g/kg/ day, gavage).The other groups were established in a mix of ways(Vit D3,high fat diet and balloon injury)while the rats of normal control group were given basic diet. Provide the groups with high-fat feeding after an intraperitoneal injection of Vit D3, and take the operation of carotid artery intimal injury four weeks later. After the operation the high-fat diet continued until the end of the tenth weeks. Take blood and vessel from arteria carotis communis in order to make a paraffin section, which was followed by routine HE staining and the observation of morphological differences between all the groups. Measure out the content of nitric oxide(NO), the activity of superoxide dismutase(SOD) and the level of malondialdehyde(MDA) in both plasma and arteria carotis communis. Measure out the content of plasma TC, TG, HDL, LDL,HDL-C via the ELISA method, and calculate the atherosclerosis index(AI). Immunohistochemistry was used to detect the expression changes of the lectin-like oxidized low-density lipoprotein receptor-1(LOX-1)in arteria carotis communis.Results:1. Compared with the content of NO(23.23±3)(36,6.22±0.65) 、 SOD(56.54±4.51)(48.43±4.43)、MDA(1.31±0.23)(4.32±0.23)in both of plasma and arteria carotis communis in the normal control group, the content of NO(7.76±0.89)(2.36±0.57)and the activity of SOD(17.65±2.19)(23.32±3.47)decreased significantly while the MDA(4.38±0.33)(7.33±0.57) level increased in the AS model group(P<0.05). Compared with the AS model group, the expression value of NO(7.65±0.67)(15.63±1.35) and SOD(21.04±2.27)(32.43±4.31)increased while the level of MDA(3.39±0.77)(2.57±0.46)decreased remarkably in the plasma in both of the low and the high dose LBP-Ⅲ group(P<0.05).In the arteria carotis communis the content of NO(3.34±0.35)(5.44±0.45)and SOD(27.67±2.18)(38.66±3.76)in the low and high dose LBP-Ⅲ group groups were apparently higher while the level of MDA(6.76±0.68)(5.46±0.78)were lower than that of the AS model group with significant differences(P<0.05).2. Compared with the content of TC(0.72±0.09μmol/L)、TG(1.13±0.09μmol/L)、HDL(0.42±0.08μmol/L)、LDL(0.13±0.03μmol/L)in the plasma in the normal control group,the content of TC(1.13±0.16μmol/L) 、 TG(1.82±0.12μmol/L) 、 LDL(0.28±0.05μmol/L)decreased significantly while the HDL(0.27±0.05μmol/L)increased in the AI modle group(P<0.05).Compared with the AS modle group,the content of TC(0.45±0.04μmol/L)(0.45±0.05μmol/L) 、 TG(0.96±0.06μmol/L)(0.87±0.06μmol/L) 、 LDL(0.15±0.05μmol/L)(0.16±0.03μmol/L)were significantly lower,while the content of HDL(0.36±0.06μmol/L)(0.45±0.07μmol/L)increased in both of the low and high dose LBP-Ⅲ group(P<0.05).Compared with the normal control group(AI 0.09±0.02),the AI of the AS modle group(AI4.40±0.16)was significantly higher(P<0.05);Compare with the AS modle group,the AI(0.40±0.19, 0.14±0.11)in both of the low and high dose LBP-Ⅲ group were significantly lower(P<0.05).3. The observation of the paraffin sections of HE staining under light microscope showed that, of the AS model group the vessel wall thickened and enlarged obviously, and the vascular seemed smooth and complete in the normal control group. The vascular endothelial didn’t thicken apparently and the muscular layer of endothelial was arranged regularly in the LBP-Ⅲ groups.4. The results of Immunohistochemistry showed that there was almost no expression of the lox-1 in the vascular tissue of arteria carotis in the normal control group while the expression of the lox-1 enhanced obviously in the arteria carotis communis of atherosclerotic model group, and the expression of the lox-1 in the LBP-Ⅲ groups was significantly less than that of the AS model group.Conclusions:LBP-Ⅲ can lower the blood fat in the process of AS in the rats; LBP-Ⅲ can increase the content of NO and SOD and decrease the level of MDA in the diseased tissue and the blood of the rats with AS. It showed that the antioxidant ability of the body was increased;LBP-III can lower the AS index of the rats and improve the form of the tissue of the rats with AS. It showed that LBP-III played a role in preventing the forming of AS; The anti-AS mechanism of LBP-Ⅲ may be related to the decreasing of the blood fat, the reduction of the oxidized damage and the decreasing of the expression level of the LOX-1.
