Objective:To study the biological effects of various zinc finger protein 750(ZNF750) mutants on ESCC.Methods:Endogenous ZNF750 was stably knocked down in KYSE150 and KYSE140 cells,then transfected with p LV-EGFP(2A)Puro vector carrying ZNF750 wild-type or mutants.Immunofluorescence method was used to observe the location of wild type ZNF750 and ZNF750 mutants. Traditonal MTT assay, colony formation assay and transwell chamber invasion assay were performed to observe the biological impact of ZNF750 mutants compared with wild type ZNF750.In addition, PCR-array was used to screen for ZNF750 related genes.Results:1. Wild-type ZNF750 located in the nucleus and the mutant S70 X located in cytoplasm. Their intracellular localization are obviously different.2. The effect of wild-type ZNF750 inhibiting cell growth, migration, and invasion was changed by ZNF750 S70 X mutant.3. There was significant difference between wild-type ZNF750 and ZNF750 S70 X in regulating the related gene expression.Conclusion:S70X mutant change biological function of wild type ZNF750, which plays the reverse role in contrast to wild type ZNF750.The mutant can promote malignant transformation and tumorgenesis in ESCC.
