Objective:Detection of temperature sensitive receptor channel TRPM4 whether expressed in human articular cartilage, discussing their normal articular cartilage in different age and different degree of articular cartilage on the change trend of osteoarthritis patients,research TRPM4 changes in articular cartilage degeneration.Methods:Collect the second hospital of shanxi medical university orthopaedic trauma amputees normal knee joint cartilage, divided into 20 to 29 age group, according to the age of 30 to 39 years old group, group of 40 ~ 49 years old, 6 cases in each group.Collection OA articular cartilage in patients with knee joint replacement, according to the extent of damage is divided into cartilage damage light group and the reorganization of cartilage damage, 6 cases in each group. ’s normal red O staining results evaluation of different age patients with knee osteoarthritis and cartilage degeneration, real-time fluorescent quantitative PCR method TRPM4 m RNA at the genetic level changes, TRPM4 protein expression level of the western blot analysis,immunofluorescence and immunohistochemical analysis TRPM4 in articular cartilage and distribution on the surface of the cell.Results:Safranin O staining showed that with the increase of patients age, gradually by smooth become coarse, articular cartilage surface area between cells staining gradually changed from deep red to pale red, OA patients sugar amino polysaccharide changes obviously. TRPM4 protein channel on knee joint cartilage. With the increase of age, each one in normal cartilage TRPM4 m RNA and protein increased gradually,the difference was statistically significant(P < 0.05), and the age and TRPM4 protein expression was positively correlated(r = 0.632, P < 0.01). With the destruction of articular cartilage, cartilage TRPM4 m RNA and protein increased significantly, the difference was statistically significant(P < 0.05), and TRPM4 protein expression and the damage degree of positive correlation(r = 0.714, P < 0.01). Immunofluorescence showed that TRPM4 mainly in the cartilage cell membranes. Immunohistochemistry showed that shallow middle TRPM4 located in cartilage.Conclusions:TRPM4 can be expressed in the human knee joint cartilage cells, exists in the light of articular cartilage cells on the membrane of middle. In the general population,as the growth of the age, the expression of TRPM4 quantity increase gradually, in patients with osteoarthritis, as the damage is aggravating, TRPM4 expression quantity increase gradually, and TRPM4 expression quantity and age change and osteoarthritis cartilage damage were positively correlated. At present, in the cartilage cells TRPM4 physiological functions and pathological physiological mechanisms are still unclear,we further research will focus on TRPM4 channel proteins in normal cartilage cells and function of arthritic cartilage cells, so that the protection of the articular cartilage degeneration and prevention, provide new targets for the treatment of osteoarthritis.
