Objective:To study the effects of serotonin at different concentrations on differentiation and of cultured primary rat osteoblasts in vitro, and to research the connection between serotonin and the loss of bone to treat the osteoporosis. Methods:Osteoblasts from the skull of newborn SD rats were divided into four groups by the culture medium with different serotonin concentrations:0mol/L group(blank cont rol),10-9mol/L group,10-8mol/L group and 10-7mol/L group. We had previously removed the serotonin of the culture medium. In addition, m RNA expression of csteocalcio(OCN), alkaline phosphatase(ALP), runt-related transcription factor2(Runx2),Osterix and Lrp5 were quantitated by real-time reverse transcription polymerase chain reaction. Results:Different concentrations of serotonin could inhibit the m RNA expression of OCN,ALP,Runx2,Osterix and Lrp5 of osteoblasts.Moreover, the inhibitory effect of serotonin was dose-dependent. Conclusions:Serotonin can inhibit differentiation of osteoblasts,which provide a novel approach for the treatments of osteoporosis.
