Objective:To prove the HIF- 1 is involved in autophagy of cardiac cells in an oxygen-free environment by observing the changes of proteins relating to autophagy of a kind of cardiac cells H9C2 in the six hours’ presence of hypoxia. Methods:1 Grouping1.1 normal control group: The cells were grown in 10% high-glucose DMEM supplement at 37℃ with a 5% CO2 without any other action.1.2 hypoxia group: the cells were grown in sugar-free and serum-free medium with hypoxia and without agents.1.3 3-MA autophagy inhibitor group: The cells were grown in in 10% high-glucose DMEM supplement at 37℃ with a 5% CO2 and 3-MA(inhibitor of autophagy).1.4 hypoxia and 3-MA group:The cells were grown in sugar-free and serum-free medium with hypoxia and 3-MA(inhibitor of autophagy).2 Methods: Werstern blot was performed to identify the protein expressed. Results:hrough comparing the changes of LC3, m TOR, Bcl-2/Bax and HIF-1 relating to H9C2 in the six hours’ presence of hypoxia among normal group, anoxia group, 3-MA group and anoxia and 3-MA group, it was found that the autophagy decreased and the HIF-1 expressed increased, which resulted in the inference that there is a connection between HIF-1 and the decreasing of autophagy. And the analysis of SPSS17.0 showed that the comparison between groups has statistical meaning after intergroup comparisons(P<0.05). It can be included that the HIF- 1 is involved in autophagy of cardiac cells in an oxygen-free environment. In conclusion:1 LC3 II / I at least six hours of hypoxia, while m TOR increased autophagy prove significantly decreased in six hours.2 Hypoxia Bcl-2 / Bax ratio decreased to prove autophagy reduce, increase apoptosis. Autophagy of cells play a protective role.3 HIF-1α hypoxia increased, since the bite inhibition of change, considering changes in HIF-1α is associated with reduced autophagy, and is located upstream.
