Objective:Used bone marrow mononuclear cells(BMMNC) from patients with chronic myelogenous leukemia(CML) as cell model to investigate the effect of interferon-( IFN-γ) on tumor necrosis factor related apoptosis inducing ligand(Tumor Necrosis factor-related apoptosis-inducing ligand, TRAIL) inducing apoptosis of BMMNC from CML patients, and to explore its mechanism of action.Methods:(1) TRAIL, IFN-γ and IFN-γ function after 24 h and BMMNC apoptosis rate set by the joint action of TRAIL detected by flow cytometry.(2) The expressions of membrane-bound TRAIL by Flowcytometry after IFN-γplaying its role 0, 6, 12 and 24 hour respectively.(3) The expressions of membrane-bound the receptor DR5 of TRAIL protein by Flowcytometry after IFN-γ playing its role 0, 6, 12 and 24 hour respectively.Results:(1) The apoptosis rate of BMMNC in group of CML control, TRAIL, IFN-γ and TRAIL+IFN-γ were(10.49 + 0.59)%,(28.50 + 3.63)%,(35.51 + 1.81)%,(54.58 +3.04)% respectively detected by Flowcytometry too. The difference was statistically significant in the group of TRAIL+IFN-γ compared with the group of CML control,TRAIL, IFN-γ(P < 0.05).(2) The mean intensity of fluorescence of BMMNC membrane-bound the average fluorescence intensity of TRAIL were(18.20 + 0.82),(22.38 + 0.90),(34.97+ 2.19),(67.29 + 2.19),respectively detected by Flowcytometry after the role of IFN-γ 0, 6, 12 and 24 h.The difference was statistically significant in the role ofIFN-γ 6, 12 and 24 h compared with IFN-γ 0h(P < 0.05). And longer duration of action the expression quantity increased further.(3) The mean intensity of fluorescence of BMMNC membrane-bound the average fluorescence intensity of its receptor DR5 were(12.70 + 1.24),(18.70 +1.62),(26.44 + 0.80),(51.93 + 1.80) respectively detected by Flowcytometry after the role of IFN-γ 0, 6, 12 and 24 h.The difference was statistically significant in the role of IFN-γ 6, 12 and 24 h compared with IFN-γ 0h(P < 0.05). And longer duration of action the expression quantity increased further.Conclusion:The combined effect of IFN-γ and TRAIL was that it could significantly increase the apoptosis of BMMNC in patients with CML, and its mechanism mifht be related to the expression TRAIL and its receptor DR5 protein up-regulated by IFN-γ.
