Objective:Under the condition of the change of oxygen,to investigate the effects of gefitinib on the proliferation of non-small cell lung cancer(NSCLC)cell line A549,HCC827, and SKMES-1.And to explore the underlying molecular mechanism.Methods:Human NSCLCA549,HCC827,SK-MES-1cells were cultured in vitro.Cells were divided randomly into the groups,after cultured for 1 day.The proliferation of these cells were investigated by CCK-8 assay,and application of real-time fluorescent quantitative PCR to detect alpha HIF-1m RNA,VEGF-C m RNA,EGFR m RNA expression.Results:CCK-8 assay results showed that under the condition of oxygen,low oxygen and high oxygen,cell proliferation inhibition rate of the treatment with the increase of the concentration and increases with the rising of oxygen concentration(P<0.001).Under the condition of high oxygen,related to the lack of oxygen of HIF-1alpha m RNA in these cell groups were significantly reduced(P<0.05),while VEGF-C m RNA 、 EGFR m RNA in HCC827 cell groups has statistical significance.Conclusions:Gefitinib can significantly inhibit tumor cell proliferation,and Enhanced with the increase of oxygen concentration,the possible mechanism is to improve the oxygen concentration can be further lowered the activation of HIF-1alpha.
