Objective:To investigate the association between Bsm I restriction site polymorphism in Vitamin D receptor gene and diabetic nephmpathy、25-hydroxy vitamin D3. Methods:PCR-restriction fragment length polymorphism(PCR-RFLP) and agarose gel electrophoresis were used to test the genotype and allele frequency of Bsm I in 150 patients with type 2 diabetic nephropathy(DN+ group)and 150 patients with non nephropathy in type 2 diabetes(DN- group)and 150 healthy individuals(NC group). Enzyme-linked immunosorbent assay(Elisa) was used to test serum 25-OHD3. Test relevant clinical and biochemical parameters at the same time.To compare the difference of VDR genotype and allele frequencies in different groups and the difference of serum 25-OHD3 in different genotypes.The DM group was divided into groups of diabetic nephropathy(DN+ group, 150 cases), non-diabetic nephropathy group(150 cases DN- group) according to urinary albumin excretion rate(UAER). Result:1. The frequency of BB+Bb genotype significant higher in DN+ group than those in DN- and NC groups(χ2=7.139,p=0.005;χ2=12.329,p<0.001);The frequency of BB+Bb genotype in DN- group with no significant difference in NC group(χ2=0.75,p=0.235).2. The frequency of B allele significant higher in DN+ group than those in DN- and NC groups(χ2=5.941,p=0.009;χ2=10.039,p=0.001);The frequency of B allele in DNgroup with no significant difference in NC group(χ2=0.545,p=0.259).3. The level of 25-hydroxy vitamin D3 significantly lower in BB group than those in Bb and bb groups(p=0.024);The level of 25-hydroxy vitamin D3 in Bb group with no significant different in bb group(p=0.147).4. Risk analysis between allele and DN showed that:odds ratio of B allele for DN was 6.507,greater that 1,95% confidence intervals(95%CI) odds ratios was 1.404-30.164,it seemed that B allele was a risk factor in patients with DN,the danger of carrying the B allele of DM in patients with DN was higher 5.507 times than untreated carry the gene.5. There is a correlation between vitamin D and diabetic nephropathy. Conclusions:1. Vitamin D not only prevent diabetes can also prevent kidney of diabetic.2. Vitamin D associated with diabetic nephropathy.
