Objective:By using RT-PCR detection of PBMCS P2X7 receptor expression between asthmatic children and non asthmatic children with RSV infection,carry out discussion on differences and significance about P2X7 receptor expression between asthmatic children and non asthmatic children(control group).Research object:Choose hospitalized children in 3 mouths to 3 years old from 2014 June to 2015 March in the people’s Hospital of Changzhi city, cases are in line with the seventh edition of "Pediatrics" children with respiratory diseases(asthma excluded)diagnostic criteria,determined by direct immunofluorescent method detect respiratory syncytial virus positive.Excluding cardiogenic asthma, tuberculosis,pertussis,bronchial foreign bodies and other large airway obstruction and congenital malformations caused by wheezing. Before entering the group nearly two weeks no use of glucocorticoid.Selected children who saticficed our standard were divided into two groups:each group with thirty children,the differences of the two group is one team with wheezing and another one without wheezing but both of with respiratory syncytial virus infection.Two groups of gender,age had no significant difference.Materials and methods:RSV positive wheezing and no wheezing symptoms in children with 30 cases in each group were collected the fresh peripheral blood,after centrifugation and precipitationextracted PBMCS(peripheral blood mononuclear cells),extracted RNA by RNAiso Plus reagent,RNA electrophoresis extraction results,measured RNA concentration of UV spectrophotometry,c DNA synthesis reaction according to the Prime Script TM 1st Strand c DNA II Synthes manual,PCR method was used to detect the expression of P2X7 receptor,the β-Actin c DNA amplification as internal reference gene sequences of primers were designed by Primer5.0.Results:By RT-PCR assay, asthmatic children’s P2X7 receptor m RNA expression levels is1.19±0.35 according to gel imaging system photo analysis,while the control group P2X7 receptor m RNA expression levels of 0.56±0.15,The difference between the asthmatic group and the non asthmatic group was statistically significant,using F test calculate the difference.α=0.05 is the test level.Conclusions:There is a statistically significant increase in P2X7 receptor expression for RSV infected asthmatic children group than non asthmatic children group. The fact that P2X7 receptors are involving in the occurrence of asthmatic symptoms in RSV infection children.
