Objective: As one of the members of the family of Wnt, WIF-1 affect the gene expression in the cell by combining Wnt protein to suppress the conduction of Wnt signaling pathways. In this experiment, we will explore the relationship between clinical pathological osteosarcoma by detecting osteosarcoma WIF-1 promoter region methylation and m RNA expression level, providing the theoretical basis for the clinical treatment of patients with osteosarcoma. Methods: 40 paraffin embedded osteosarcoma tissues and 20 matched paraffin embedded tissues of chondroma was procured from the Affiliated Hospital of Qingdao University after approval from2008 to 2014 and procured hospitalized and pathological diagnosis information. By using methylation specific PCR technology(methylation specific PCR, MSP) detection of osteosarcoma tissue and chondroma tissue Wl F-1 gene promoter region methylation status, research Wl F-1 gene methylation status and the correlation between clinical pathology. Application of real-time quantitative RT-PCR technique to detect WIF-1 gene m RNA in osteosarcoma and chondroma tissue expression level, research the expression of m RNA and the correlation between clinical pathology.In summary, we analyze the osteosarcoma WIF- 1 gene methylation rate and the relationship between the relative expression of m RNA, to explore the role of both in the occurrence and development process of osteosarcoma. Using SPSSl9.0 statistical software for all data statistics processing, using X2 test or Fisher exact probability method, t test and other statistical methods, the difference is statistically significant(p<0.05).Results: Wl F-1 gene in osteosarcoma tissues methylation rate was 55%(22/40) significantly higher than osteochondroma tissues methylation rate 20%(4/20)(P < 0.05).WIF-1 gene methylation positive was not correlated with age, gender, etc(P > 0.05), but in the pathology of low differentiation osteosarcoma group methylation accounted for 82.61%(19/23) was significantly higher proportion of unmethylation osteosarcoma group 17.65%(3/17)(P < 0.05). WIF-1 m RNA expression in the group of osteosarcoma and osteochondroma were(0.361 ± 0.303) and(0.760±0.256)(P < 0.001).WIF- 1 gene m RNA expression was not correlated with age, gender, etc(P > 0.05), but in pathologic low differentiation group was(0.201±0.233) significantly lower than the high differentiation group(0.578±0.250)(P < 0.001). The WIF-1 m RNA expression in the WIF-1 promoter region methylation status of osteosarcoma(0.139±0.131)significantly lower the methylation status(0.642±0.188)(P < 0.001). Conclusion: WIF-1 gene promoter region methylation may be abnormal activation of the classic and the classic Wnt signaling pathways that affect m RNA expression, which affects the development of osteosarcoma.
