The occurrence of tumor is a complex process based upon a sequence of interactions between endothelial cells and tumor cells. Most of the former research about co-culture with endothelial cells and tumor cells focused on stimulation from tumor cells to vascularization and the expression of angiogenesis related factor, but little is known about the malignant impact of vascular endothelial cells to tumor cells, and stay in the two-dimensional level.To model tumor/endothelial-cell interactions, we bulit the co-culture system for breast cancer cells(4T-1) and human umbilical vein endothelial cells(HUVECs).we used biomaterial including sodium alginate, gelatin, chitosan and hyaluronic acid to built hydrogels. In this study, we conside many factors, including cell percentage,the total number, different cultivation methods and the environment in co-culture system. Analysing of tumor cell growth, we built the suitable co-culture system for4T1 with HUVECs in the three-dimensional environment.We mainly studied reaction of tumor cells under the action of endothelial cells in the aspects of stem cell properties(CD44, CD24), invasive migration ability(MMP9)and multi-drug resistance(MDR1). The results show that endothelial cells directly impact on the growth, proliferation, migration and other malignant of tumor, played crucial roles in RNA expression of tumor cells in vitro, consistenting with the result of protein detection.In the last part, we studied interaction of cancer stem cells and breast cancer cells by co-culture system. Our data suggest that significantly promoting effect from HUVECs to 4T1-CSC. Consistent with immunofluorescence, RT-PCR of expressed m RNA revealed the varying upregulation of gene expression with MMP9, MDR1,CD44, Sca1, Nanog and Sox2 for 4T1-CSC in co-culture system.These results demonstrated that co-culture of HUVECs and 4T1 changed cell biological characteristics. Based on this knowledge, the interaction between tumor cells and endothelial cells have close relationship, as the target of tumor therapy.
