The infection of polio-virus can cause a cute intestinal infectious disease, with the main symptoms including acute flaccid paralysis, lower limb hypoplasia and dysfunction, even permanent disability. The main preventive method in epidemiology is the use of Oral Polio Vaccine(OPV) and Inactive Polio Vaccine(IPV). However, the Vaccine associated paralytic poliomyelitis(VAPP) and vaccine-derived polioviruses(VDPV) are found in the countries which are using OPV, suggesting the control of the vaccine quality require to be improved, so is the method in rapid screening the suspected polio cases.This study aims to build three types of armored RNAs, Armored RNA-sab I, Armored RNA-sab II and Armored RNA- sab III, and to set the armored RNAs as a positive control in the real-time fluorescent quantitative polymerase chain reaction(q RT-PCR).In this study we use PAR engineering plasmid as the carrier, connected to the specific gene of polio virus strains sabin I, sabin II and sabin III to get combination plasmids PAR-sab I, PAR-sab II and PAR-sab III, then transformed into the competent Escherichia. The recombined strains were cultivated and induced, then centrifuge and add enough DNase I for the primary Armored RNAs. After detected, Armored RNAs can be used in q RT- PCR to obtain standard curve and for further acknowledge.
