Objective: To investigate effects of CADM1 overexpression on proliferation,invasion and metastasis of human osteosarcoma cell line U-2 OS, and its relationship with ERK5 signaling pathway.Methods: 1. Detecting the different expression between U-2 OS and h FOB 1.19 by using Western Blot. 2. Making CADM1 overexpression in the cell line U-2 OS by transfection. 3. Proving effects of CADM1 overexpression on proliferation, invasion and metastasis of human osteosarcoma cell line U-2 OS by MTS,Transwell and Wound healing. 4. Confirming the relationship between ERK5 signaling pathway and CADM1,s effects which contain suppressions of proliferation, invasion and metastasis by adding BIX02188.Results:1. The expression of CADM1 in human osteosarcoma cell line U-2 OS is lower than h FOB 1.19. 2. Transfection raised the C ADM1 expression in human osteosarcoma cell line U-2 OS by 20 times. 3. In MTS, Transwell and Wound healing, we found that CADM1 can impress proliferation, invasion and metastasis of human osteosarcoma cell line U-2 OS. 4. Blocking the ERK5 signaling pathway, proliferation, invasion and metastasis were impressed in LP-NEG-Lv201-100 U-2 OS,by contrast,they increased in LP-S0406-Lv201-400 U-2 OS.Conclusion: 1.The expression of CADM1 is reduced obviously in the human osteosarcoma cell line U-2 OS 2. CADM1 can impress the proliferation, invasion and metastasis of human osteosarcoma cell line U-2 OS. 3.ERK5 signaling pathway takes part in the process of CADM1 impressing human osteosarcoma cell line U-2 OS.
