The Expression And Clinical Significance Of HCCR, Survivin And PTEN In Human Gastric Cancer

BACKGROUND:Cancer of the stomach(gastric cancer) is one of the most common malignant tumor in the world. In our country, the cancer of stomach are multi-stage, multi- step and orderly process and it are comprehensive result of many fac tors, including tumor suppressor genes, oncogenes, and some growth factors which play an important role in the developing of stomach cancer. OBJECTIVE:Observe the expression of HCCR, PTEN and Survivin in gastric cancer tissues, adjacent tissues and normal gastric tissues, and explore the relationship between them and the clinicopathological features of gastric cancer。Para carcinoma and normal gastric tissues. In addition, we are going to detect the expression of PTEN,Survivin and HCCR in the occurrence and develop me of its molecular mechanism, whose we can understand the cancelation of signaling pathways and provide exploration experiment on molecular target therapy and the basis of clinical treatment for gastric cancer. METHOD:1: we have collected normal gastric, Para-carcinoma and gastric cancer tissues 60 cases respectivly,in the period of 2012.11-2013.11 in people’s Hospital of Gan Su.2:All of the specimens of patients selected have not receive radiation therapy, chemotherapy and others targeted therapies. Furthermore, the specimens selected uncoincidence other tumors. 3: Using immunohistochemical S-P method to detect the expression of HCCR, Survivin and PTEN in ten gastric cancer, adjacent cancer, normal gastric mucosa tissues 60 cases respectively. Using SPSS22.0 statistical software to statistics all the data, the methods are using chi-square tests and Spearman correlation analysis which will be own differences significant under P<0.05. RESULT:HCCR express 2 cases only in normal gastric tissue of 60 cases and the expression rate are 3.3%. Besides, it have 27 cases masculine gender in 60 cases Para-carcinoma tissues that expression rate are 45% and 43 cases masculine gender in 60 cases gastric cancer tissue whose expression rate are 71.7%. Its positive expression rate have the increasing trend from normal gastric and para-carcinoma tissues to gastric cancer tissues. HCCR are expression in para-carcinoma and gastric cancer tissues significantly higher than norma stomach tissue which the discrepancy has the significance extremely significant(P<0.05); para-carcinoma and gastric cancer tissues express also have the significance extremely significant(P<0.05)PTEN have 58, 47 and 28 cases positive expression in 60 cases normal gastric, adjacent of cancer and gastric cancer tissues which the expression ratio are 96.7%, 78.3% and 46.7% respectivly. In normal gastric, adjacent of cancer and gastric cancer tissues, the PTEN positive expression ratio have a decline trend. More, it express lower in adjacent of cancer and gastric cancer tissues than normal gastric tissues that the discrepancy have significant sense(P<0.01). At the same time, it expresses in adjacent of cancer and gastric cancer tissues also have significant sense(P<0.05).Survivin have 7, 26 and 44 cases positive expression in 60 cases normal gastric, adjacent of cancer and gastric cancer tissues which the expression ratio are 11.7%%, 43.3% and 73.3% respectivly. In normal gastric, adjacent of cancer and gastric cancer tissues, the Survivin positive expression ratio have the increasing trend obviously. Furthermore, it expresses high in adjacent of cancer and gastric cancer tissues this discrepancy have significant sense(P<0.01). In adjacent of cancer and gastric cancer tissues, it also have visible difference(P<0.05)The HCCR and Survivin gene express clearly in gastric cancer tissue. But, the PTEN protein expression low which has the statistics significant(P<0.05); the express of Survivin gene has relationship with pathological level in gastric cancer tissue(P<0.05).CONCLUSION:HCCR and Survivin gene protein are expression high in gastric cancer tissues, which provides a new biological indicator for diagnosis gastric cancer early. At the same time, it provides the new treatment strategies. PTEN gene express lower and lack relatively in gastric cancer tissue has the relationship with stomach cancer take place, whose may provide a new clinical evaluating on tumor malignant degree and prognosis.
