The Effect Of Banxiaxiexin Soup Medicated Serum On Proliferation And Invasion To The Peritoneal Metastasis Gastric Cancer Cellular

Objective:through the Banxiaxiexin soup medicated serum intervention in human gastric cancer peritoneal metastasis GC9811-P cells,to study the GC9811-P cells’ growth,migration,apoptosis and TGF-beta1,VEGFm RNA and protein expression level after the tumor micro-environment being induced.Discussing Banxiaiexin soup intervention mechanism of peritoneal metastasis of gastric cancer,provide experimental basis for traditional Chinese medicine treatment of tumor.Methods:four rabbits were respectively irrigated Banxiaxiexin soup and physiological saline,getting blood from the hearts in sterile conditions.Mixture the medicated serum into the concentration of 25μL/m L,50μL/m L and100μL/m L to intervene the GC9811-P cells.With RTCA testing the cells’ growth and migration.Flow cytometric method to detect cell apoptosis and cell cycle.Observe the cellular morphology and mitochondrial membrane potential who was dyed by rhodamine123 under the laser confocal microscope.RT-PCR detect the expression of TGF-beta1 and VEGFm RNA,Western blot detect the protein expression on level.Results:according to the experimental results show that GC09811-P cells through the different concentrations of Banxiaxiexin drug-containing serum(0,25,50,100μL/m L)Intervene,the cell proliferation was significantly inhibited,and the drug-containing serum concentration was positively related with cell proliferation inhibition.RTCA(real-time dynamic detection) show that the control group(SF)didn’t migration.Others Which were intervened by Banxiaxiexin containing serum(0,3.1,6.2,12.5,25,50,100μL/m L)groups all migrate.To 23 h,migration ability has no obvious difference(P>0.5),after the 23 h,25,50,100 u L/m Lgroups could significantly inhibit the migration ability,and the concentration dependence.Compared with group1(blang group),group2(25μL/m L),3(50μL/m L),4(100μL/m L)of the percentage of cells in G2/M phase,S lower,the effect relative with the dosage.The laser confocal microscope results show compared with the control group,the concentration of each group of intracelluar fluorescence intensity isabate,mitochondrial membrane potential is reduced,and the concentration is higher,the intensity is weaker,the membrane potential is lower.RT-PCR and Western blot results show that the Banxiaxiexin soup medicated serum can down regulation VEGFm RNA,with drug-containing serum concentration increases,TGF-beta1 and VEGF protein expression decreased,and were positively correlated.Conclusion:Banxiaxiexin soup medicated serum can inhibit the human gastric cancer peritoneal metastasis GC9811-P cell proliferation and migration and its mechanism may be related to inhibition of TGF-beta1 and the secretion of VEGF.
