Objective:To investigate the correlation of CYP11B2 and CYP11B1 and the formation of aldosterone-producing adenomas(APA) by detecting the expression of CYP11B2 and CYP11B1 in the adrenal tissue of patients with APA.Methods:From July 2013 to July 2014, 30 cases of adrenal placeholder with APA were collected after the adrenalectomy from the First Affiliated Hospital of Gannan Medical University and China Hospital of PLA, including 16 cases of males,14 cases of females, with an average age of 45(range 26 to 66) years. 10 cases of normal adrenal tissue from patients adrenalectomized for renal cancer were collected as the control group, including 5 cases of males and 5 cases of females, with an average age of 52(range 40 to 71) years. The expression of CYP11B2 and CYP11B1 m RNA was detected by real time RT-PCR and the expression of CYP11B2 and CYP11B1 protein was detected by Western blot. The data were analyzed by SPSS14.0 statistical software(SPSS, Chicago, IL, USA). Comparisons among groups were performed by a two-sample t-test or analysis of variance. The correlation between the expression of CYP11B2 and CYP11B1 and APA preoperative clinical features were performed by correlation analysis. The correlation between the expression of CYP11B2 and CYP11B1 m RNA and protein were performed by correlation analysis. P<0.05 denoted the difference was statistically significant.Results:The expressions of CYP11B1 and CYP11B2 gene were detected in all the groups, including ten normal adrenals and thirty APA tissues. The ΔCT of CYP11B2 m RNA in experimental group and control group were 1.46±0.93 and3.92±2.42, respectively, the difference was statistically significant(P=0.005).TheΔCT of CYP11B1 m RNA in experimental group and control group were-2.27±1.63and-2.51±2.43, respectively, the difference was no statistically significant( P=0.764).The levels of the relative expression of CYP11B2 protein in experimental group and control group were 0.6160±0.085 and-0.6606±0.143,respectively, the difference was statistically significant(P<0.01). The levels of therelative expression of CYP11B1 protein in experimental group and control group were-0.7866±0.054 and-0.2509±0.092, respectively, the difference was statistically significant(P<0.01). In APA, the relative expression of CYP11B2 m RNA are positive correlated with the level of aldosterone(r =-0.629, P<0.01) and negative correlated with the level of potassium(r = 0.410, P=0.025); the relative expression of CYP11B1 m RNA are negative correlated with preoperative highest systolic blood pressure(r=0.364, P=0.048); the expression of CYP11B1 protein are negative correlated with age(r =-0.385, P=0.036); the expression of CYP11B2 protein are negative correlated with the expression of CYP11B1 m RNA(r =-0.361, P=0.050).Conclusions: The overexpression of CYP11B2 gene might promote the formation of APA by influence the independent high aldosterone secretion. The overexpression of CYP11B2 protein might be correlated with the low expression of CYP11B1 gene. CYP11B1 gene might participate in in the formation of APA.
