| Objective:To explore HMGB1 〠NF-κB and Inflammasome NLRP3 〠ASC and AIM2 expression levels and the relationship with the occurrence, development and prognosis in patients with acute leukemia,it maybe provide laboratory evidence for diagnosisã€treatment and recurrence prediction of acute leukemia.Methods:We collected the blood and bone marrow samples from 18 cases of ALL and 42 cases of ANLL a total of 60 patients with acute leukemia(experimental group) and 20 patients without malignance disease(control group) in the Hematology Department of the First Affiliated Hospital of Gannan Medical College from July 2013 to July 2014.The experimental group was divided into untreated groupã€recrudescent group and complete remission group,each group was 20 cases.ELISA detect HMGB1ã€NLRP3ã€ASCand AIM2 antigen level in plasma,RT-PCR detect HMGB1 and NF-κB gene level in bone marrow mononuclear cells,Western blot detect HMGB1 and NF-κB protein level in bone marrow mononuclear cells,IHC detect HMGB1 and NF-κB expression level in bone marrow.Results:ELISA showed that HMGB1 antigen level in untreated group and recrudescent group were significantly higher than in control group(P<0.05),NLRP3ã€ASCã€AIM2 antigen level doing the opposite,there was no statistically significant difference between complete remission group and control group(P>0.05); and there was no statistically significant difference between ALL and ANLL( P > 0.05).RT-PCR showed that HMGB1 and NF-κB gene expression were significantly increased in untreated group and recrudes-cent group compared to control group(P<0.05),but not in the complete remission group(P>0.05),there was no statistically significant difference between ALL and ANLL(P>0.05).Western blot showed that HMGB1 and NF-κB protein expression levels were significantly elevated in untreated group and recrudescent group(P<0.05), but not in complete remission group(P>0.05),there was no statistically significant difference between ALL and ANLL( P >0.05).IHC showed that HMGB1 and NF-κB is mainly expressed in untreated group and recrudescent group.HMGB1 was positively correlated with NF-κB,but negatively correlated with NLRP3 〠ASC and AIM2.The survival time of HMGB1 overexpression patients is longly than that of non-overexpression patients.Conclusions:1. The Inflammasome NLRP3ã€ASC and AIM2 may have the function of anti leukemia.2. HMGB1 involved in the occurrence and development of leukemia maybe through activating the NF-κB signaling pathway.3. The expression level of HMGB1 can be a indicators to predicting relapse of leukemia.4. HMGB1 can be the prognostic factors of patients with leukemia. |
