The purpose:To investigate the function of the correlation between Th-POK and T cells differentiation in gastric carcinoma tissues. Methods:We selected 100 cases of patients received gastric cancer surgery in the first affiliated hospital of Nan Chang university from March 2011 to October 2014, then collected gastric cancer tissue and normal tissue adjacent to carcinoma, together with clinical data and pathological data. 71 of the patients were male and 29 were female; Patients aged 25-86, and the average age is 58.9±11.0 years old, 46 patients are younger than 60 years old, while 54 patients are or older than 60 years old; there are gastric fundus carcinoma in 9 cases(9%), the gastric body cancer in 29 cases(29%) and gastric antrum carcinoma in 62 cases(62%). 69 cases with lymph node metastasis while 31 cases without. There are 61 cases with high or moderate carcinoma differentiation, and 39 cases with low differentiation; According to the new TNM staging(see appendix), a total of 45 cases are of stage TNM staging I-II, and 55 cases are in stage III and IV. Depending on immunohistochemical method,we detected Th POK expression in lymphocytes of gastric cancer tissue and normal tissue adjacent to carcinoma, as well as the the expression of CD4, CD8, RUNX3, GATA3, GZMB protein. Results:1, Th POK positive expression rate is higher in lymphocytes of gastric cancer tissue than tissue adjacent to carcinoma(80% vs 67%, P<0.05), meanwhile the intensity of positive expression is significantly higher than tissue adjacent to carcinoma(P<0.01). Of the patients in TNM staging Ⅲ-Ⅳ, Th POK expression intensity in gastric cancer tissue were significantly stronger than of patients in TNM staging Ⅰ-Ⅱ(P < 0.05).2, CD4 positive expression rate is higher in lymphocytes of gastric cancer tissue than tissue adjacent to carcinoma(77% vs 60%,P< 0.01), meanwhile the intensity of positive expression is significantly higher than tissue adjacent to carcinoma(P<0.01). Of the patients in TNM staging Ⅲ-Ⅳ, Th POK expression intensity in gastric cancer tissue were significantly stronger than of patients in TNM stagingⅠ-Ⅱ(P < 0.05).3, CD8 positive expression rate is lower in lymphocytes of gastric cancer tissue than tissue adjacent to carcinoma(80% vs 60%, P<0.05), meanwhile the intensity of positive expression is significantly lower than tissue adjacent to carcinoma(P<0.01). Of the patients in TNM stagingⅠ-Ⅱ, Th POK expression intensity in gastric cancer tissue were significantly stronger than of patients in TNM staging Ⅲ-Ⅳ(P < 0.05).4、RUNX-3 positive expression rate is significantly lower in lymphocytes of gastric cancer tissue than tissue adjacent to carcinoma(59% vs 86%, P<0.01), meanwhile the intensity of positive expression is significantly lower than tissue adjacent to carcinoma(P<0.01). Of the patients in TNM stagingⅠ-Ⅱ, Th POK expression intensity in gastric cancer tissue were significantly stronger than of patients in TNM staging Ⅲ-Ⅳ(P < 0.05).5, GATA3 positive expression rate is higher in lymphocytes of gastric cancer tissue than tissue adjacent to carcinoma(76% vs 62%, P<0.05), meanwhile the intensity of positive expression is significantly higher than tissue adjacent to carcinoma(P<0.01). Of the patients in TNM staging Ⅲ-Ⅳ, Th POK expression intensity in gastric cancer tissue were significantly stronger than of patients in TNM staging Ⅰ-Ⅱ(P <0.05).6, there was no significant difference of both GZMB positive expression rate and expression intensity between in gastric cancer tissue and normal tissue adjacent to carcinoma(70% vs 61%, P>0.05). Meanwhile there was no significant difference of the expression intensity of GZMB in lymphocyte in Gastric cancer tissues between each group of different pathological features.7, The positive expressiong rate and intensity of Th POK protein in lymph cells in gastric cancer have positive correlation with CD4 and GATA3 protein, and The expression of CD8 and RUNX3 showed a negative correlation with Th POK expression intensity. Conclusion:Expression of Th POK in lymph cells of gastric cancer tissue is higher than normal, by which interact with RUNX3 and GATA3 to counteract the CD8 + lymphocytes generated, increases the generation of CD4 + lymphocytes and helping to gastric cancer immune escape.
