Objective: To investigate the effects of silence Aurora-B on growth of human osteosarcoma cell line 143 B in nude mice.Methods: 143 B human osteosarcoma xenografts were used as preclinical tumor models. The osteosarcoma nude mice(18) were divided randomly into three groupsand and injected with LV-sh Aurora-B、LV- negative and PBS in tumor tissue every 3 days for 3 times, respectively.The skin measuring size of the tumor were measured every 7 days,After 5 weeks,the mice were sacrificed and the weigh and size of the tumor were measured. The apoptosis of transplanted tumor tissues were measured by the TUNEL assay.Result: The tumor growth in nude mice injection with LV-sh Aurora-B was inhibit significantly, compared with the other two groups(P<0.05). After 5 weeks, the weigh and size of the tumor injection with LV-sh Aurora-B was significantly lower than the other two groups. The apoptosis rate of transplanted tumor tissues in nude mice injection with LV-sh Aurora-B was significantly higher than that in the other two groups(P<0.05).Conclusion: Silence Aurora-B could effectively induce apoptosis and inhibit the growth of human osteosarcoma 143 B cells in nude mice.
