Objective:To explore effect of Cetuximab conjugated tubulin inhibitorin the treatment A549 cell transplantation tumor in nude mice.Method:Preliminary experiment : the CCK-8 method was used to detect the inhibitory effect of Cetuximab conjugated tubulin inhibitor and cisplatin on human lung cancer A549.To establish animal model of lung cancer A549 nude mice, 20 nude mice were randomly divided into 4 groups: blank control group, Cetuximab conjugated tubulin inhibitor group(ADC group), cisplatin group, cetuximab group. To observe the tumor growth, plot of tumor growth.using TUNEL method to detect apoptosis, then calculated cell apoptosis index(AI).Result:1.Preliminary experiment : Inhibition rate of 2.67 nmol cetuximab conjugated tubulin inhibitor(ADC 100 ul 4mg/ml) significantly higer 133 nmol cisplatin(10ul4mg/ml).The ADC molar concentration is 0.02 times of cisplatin. NOTE: cisplatin molecular weight 300.05 g / mol, Cetuximab conjugated tubulin inhibitor molecular weight of about 150000 g / mol.2. In vivo experiment:Statistics tumor volume, AI, ADC group than cetuximab,the control group were significantly different(P <0.05), cisplatin group than the other three groups were significantly different(P <0.05). Note: 2.67 nmol ADC,332.5nmol cisplatin.The ADC molar concentration is 0.008 times of cisplatin.Conclusion:Preliminary experiment:significantly inhibited of cetuximab conjugated tubulin inhibitor on lung cancer A549 cells.Cetuximab conjugated tubulin inhibitor has certain inhibition and apoptosis on huaman lung cancer A549.
