Objective 1. To explore the difference about the expression of IGF-2 between the low-grade and high-grade gliomas patients in tissue 2. To detect he expression of IGF-2 between the low-grade、high-grade gliomas patients and the normals in serum.Methodsand materials Experiment I:We collected 25 cases of gliomas specimens(12 low-grade cases and 13 high-grade cases) in Ningxia Medical University General Hospital From April to December in 2013,the tissue specimen wasclassified according to the WHO classification in 2007, and are the incipient, has not been preoperative radiotherapy and chemotherapy.Using the methods of HE, IHC and RT-PCR to observe the expression of IGF-2 in high-grade and low-grade tissue in gliomas. Application of statistical software SPSS 16.0 for data analysis. The datas of IHC were analyzed by nonparametric tests-Mann-Whitney,and the datas of PCR were dealt with Independent sample t test. Experimental datas expressed as mean±standard deviation, data using single-factor analysis of variance between groups after the homogeneity of variance test, with p ≤0.05 was considered statistically significant.Ⅱ:We collected 25 cases of gliomas patients’preoperative fasting blood samples(12 low-grade cases and 13 high-grade cases) in Ningxia Medical University General Hospital From April to December in 2013,and the control blood samples(12 cases) were collected from thenormal physical examination population of Ningxia Medical University GeneralHospital Clinical Laboratory.And to detect the expressionlevels of IGF-2 in high-grade、low grade and control group in serum by ELISA.Using the method of single factor variance analysis to analyze the experiment data,with p≤0.05 was considered statistically significant.Results Experiment I:HE:Under the Optical microscope,we can find that the cells in high-grade gliomas tissue arranged disorderly, nucleus shaped irregularly and had a deeply stainingin cytoplasm.While,the cells in low-grade gliomas tissue formed relatively neat, arranged orderly and the cytoplasm in a lighter color.IHC:The level of IGF-2 expressed in tumor of patients with high-grade group is far higher than the low-grade group by IHC(P<0.05). RT-PCR shows the same trade that the relatively quantity of the high group-grade (1.19±1.40) is higher than the low group-grade (0.06±0.02), p=0.007<0.05, the difference was statistically significant.Experiment Ⅱ:The level of IGF-2 presented in serum as follows: High-grade group (541.85± 137.45) is higher than low-grade group (423.60±141.80), and far higher than the normal group(294.50±106.23).Conclusions 1.IGF-2 plays a role in the production and procession of gliomas,the expression level has a trend of high-grade higher than the low-grade,and it is associated with the classification of gliomas.2.The detection of IGF-2 in blood shows that the expression of high-grade is higher than the low-grade、than the control,and it would be an useful factor to guide the diagnosis and therapy of the gliomas...
