This paper studied on the inhibition pulmonary fibrosis of the chemical composition in Rhodiold sachalinensis A.Bor. Five parts were as follow.Section 1 is comprised of the extraction and separation of rhodiola. The column chromatography and HSCCC were used on ethyl acetate phase to get pure compounds which were identified by MS, 1H-NMR and 13C-NMR. Ethyl gallate, salidroside and gallic acid were identified. Rrhodiola polysaccharide was extracted also.Section 2 is comprised of the extraction process optimization of salidroside. The response surface analysis was used on salidroside in rhodiola from Changbai Mountain. At last, the optimized conditions were defined as 70% ethanol concentration, 1 hour to extract, 10:1 liquid and solid ratio. The extraction rate was 1.26% by this condition.Section 3 is comprised of the inhibition pulmonary fibrosis research of rhodiola polysaccharide. Rhodiola polysaccharide was treated with TGF-β1-induced A549 cells. The results showed that rhodiola polysaccharide made cell morphology transform to normal cell, decreased cell death rate, enhanced cell proliferation vitality and reduced FN-EDA protein. It was verified that rhodiola polysaccharide could inhibit TGF-β1-induced A549 pulmonary fibrosis well.Section 4 is comprised of the inhibition pulmonary fibrosis research of salidroside. Salidroside was treated with TGF-β1-induced A549 cells. The results showed that salidroside made cell morphology transform to normal cell, decreased cell death rate, enhanced cell proliferation vitality and reduced FN-EDA protein. It was verified that salidroside could inhibit TGF-β1-induced A549 pulmonary fibrosis well.Section 5 is comprised of protection of salidroside against bleomycin-induced lung fibrosis in mice. Salidroside can inhibit their serum Hyp and TGF-β1, reduce SOD, POD, CAT and reduce IV collagen, MDA and TGF-β1 levels. As can be seen from the pathological changes in the lung tissue, inflammatory cells decreased after using salidroside significantly.The results showed that rhodiola polysaccharides and salidroside inhibited the formation of pulmonary fibrosis in vitro well. It also can be seen that salidroside had a good protective effect on bleomycin-induced mice in vivo. It indicated that rhodiola had a good job in inhibitiing pulmonary fibrosis.
