osteoporosis is a systemic diseases characterized by increased bone fragility and damaged microstructure caused by decreased bone mass and destroyed bone microstructure. Fracture is the most dangerous complication. It is the cause of why osteoporosis so important in clinic and public health practice. Clinic research find that healing of osteoporotic fracture been delayed, and have high fatality and disability. Understand how osteoporosis influence bone repair and find way to management osteoporotic fracture is of great importance. we investigate how osteoporosis impact bone repair by observe the process of intramembranous bone repair and endochondral bone repair in osteoporotic mice. Method1. Establish osteoporotic mice by glucocorticoids or ovariectomy. Use micro-CT insure establishment of osteoporosis. 2. Establish intramembranous bone repair model of normal mice and osteoporosis mice respectively. Using HE stain, immunohistochemistry and TRER stain observe the difference between osteoporosis mice and normal mice at 7, 10, 14, 21 days after surgery. 3. Establish endochondral bone repair model of normal mice and osteoporosis mice respectively. Using HE stain, immunohistochemistry and toluidine blue stain observe the difference between osteoporosis mice and normal mice at 7, 10, 14, 21 days after surgery. ResultData of micro-CT showed that subcutaneously inject meprednisone 3.5mg/kg/d 60 days or ovariectomy 60 days could induce osteoporosis of mice. Compared with normal mice, the intramembranous bone repair of osteoporosis mice delayed. The endochondral bone repair of osteoporosis mice also delayed compared with normal mice. ConclusionIn osteoporosis mice the proliferation and differentiation of stem cell impaired, so the process of intramembranous bone repair delayed. The hypertrophy of chondrocyte and the appearance of ossification center delayed is the reason why intramembranous bone repair impaired. Thought the mechanism of glucocorticoids induced osteoporosis and ovariectomy induced osteoporosis is difference, but they have the similar way to impair the intramembranous bone repair and endochondral bone repair.
