Inonotus obliquus(IO(Fr.) Pilat) belongs to the eumycota, basidiomycotina, hymenomycetes, brown fungi order, polyporaceae, and Poria hypobrunnea Petch, and in Russia it is usually called chaga. As a medical fungi, it grows in area at about northern latitude in the Northern Hemisphere. It is a kind of slightly bitter and astringent fungi with hard polyspora and usually living in forests with dark light and rather high humidity. It mainly grows at rotting trees or the roots of them.Inonotus obliquus has been well known by people and won extensive attention from scholars in all nations. It has been found through researches that there are a large amount of polysaccharide, which could inhibit the spreading of cancer cells in tumor and improve the immunity of cells so as to achieve functions like anti-cancer, antioxidant, and improvement of immunity. Polysaccharide has become a major developing direction in the development of researches on anti-tumor drugs nowadays.In the aspects like extracting methods and activity, scholars in many countries have already done a great deal of exploring researching work, but most of them has focused on the level of crude polysaccharide and the extraction of polysaccharide has also made through water digestion with microwave assistance as the major assisting methods. Below are the contents mainly studied by this article through investigating tremendous documents and in combination with the actual experimental status of this laboratory:1.Water, oxamide(3%), and alkali(5%Na OH) were taken as the solution to extract the polysaccharide from Inonotus obliquus respectively, and the method of light assistance had been applied to obtain the best process conditions for extraction: the ratio between material and liquid was 1:30, the temperature was 95℃, the extraction time was 2.5 hours, and the power of the lightwave was 1200 W. It was proved that light wave assistance could be of some effect of raising the extraction rate of crude polysaccharide from Inonotus obliquus.2.The extraction rate of polysaccharide and the content of polysaccharide were taken as the indexes for judgment, and the three influential factors of the ratio between material andliquid, extraction time and extraction temperature had been utilized to design orthogonal test so as to obtain a set of best extraction conditions for optimize the traditional method of water digestion, that is, the ratio between material and liquid is 1:30, the extraction temperature is 95℃, and the extraction time is 2.5h.3.Three different polysaccharide elements were obtained through alcohol classification, that is, IOP40, IOP60, and IOP80 with the contents as 19.4%, 41.12%, and 9.23% respectiv4.The Sevage method and macroporous resin method were taken to eliminate protein and pigment so as to further purify Inonotus obliquus polysaccharide, and the Sevage method was also optimized through orthogonal test with the optimizing conditions as that the amount of the added Sevage agent was two times of the volume of the polysaccharide extracting solution, the ratio between the volume of chloroform and that of n-butanol was4:1, the vibration and shaking time was 25 minutes, and the deproteinization was conducted for three times.5.Three water-soluble polysaccharide fractions(IOP40, IOP60 and IOP80) were obtained and their physicochemical properties,including total sugar content, protein content, monosaccharide composition and percentage were analyzed.6.The functions of antioxidant activity in vitro of IOP40, IOP60, and IOP80 had been studied according to the test results on and the cleaning ability of 2, 2 PCB-1-the bitter taste, hydroxyl radical, superoxide anion, and hydrogen peroxide. Generally speaking, with the rising of the concentration within the scope of test, the antioxidant capacity of the three kinds of polysaccharide elements has been gradually enhanced. As to the order of the capacities of reducing power, DPPH scavenging activity, H2O2 scavenging activity, and the scavenging activity of hydroxyl radical, it was always IOP60 > IOP40 > IOP80. All these findings have showed that the three kinds of polysaccharide elements extracted from Inonotus obliquus, especially IOP60, can be used as natural elements to make function foods and in medical industry so as to relieve the pressure of oxidation.
