Objective: By constructing the rat obstructive sleep apnea syndrome(OSAS) and complicated hypertension model, this research aims to discuss the apoptosis mechanism of hippocampal neurons, providing the theoretical basis for primary prevention or clinical treatment of brain stroke in the future.Methods: 20 adult male SD rats and 10 adult male SHRs(spontaneously hypertensive rats) were divided into normal control group, OSAS model group, and OSAS complicated hypertension model group respectively, and each of these groups contained 10 rats; after the intraperitoneal anesthesia, several parts of pharyngopalatine arch and root of tongue were injected with sodium hyaluronate gel until those rats suffered apnea; after 2-4 weeks of the model construction and feeding, the electroencephalogram(EEG) of rats, saturation of blood sample and mouth, nose airflow were detected under the multi-parameter nerve monitor to determine if OSAS model was successfully constructed or not; the blood pressure of rats was dynamically monitored through the tail-artery blood pressure measurement; the brain hippocampus was taken after 14 weeks of feeding, the expression of hippocampus Caspase-3; and the expression of hippocampus Bcl-2 were detected using the western blotting method(WB) and immunohistochemical method respectively.Results: 1. The relative optical densities of WB band are: normal control group(0.19±0.09), OSAS group(0.73±0.06), OSAS complicated hypertension group(0.77±0.07); by comparing OSAS group, OSAS complicated hypertension group and normal control group, P<0.05, suggesting the statistical significance; 2. The number of Bcl-2 protein positive cells are: normal control group(104.34±12.34), OSAS group(155.38±28.36), OSAS complicated hypertension group(178.65±26.68); by comparing OSAS group, OSAS complicated hypertension group and normal control group, Bcl-2 positive rate significantly increases, P<0.05, suggesting the statistical significance.Conclusion: 1. In the apoptosis mechanism of OSAS hippocampal neurons, OSAS has the synergistic effect with associated factors of hypertension; 2. The protein expression of OSAS rat hippocampal Caspase-3 increases; 3. The expression of OSAS rat hippocampal Bcl-2 increases.
