| Objective: To examine the expressions and correlations of RUNX3, Jagged1, E-cadherin and N-cadherin in normal gastric mucosa, precancerous lesions and gastric cancer tissues.Methods: Tissue samples of grastric precancerous lesions(chronic atrophic gastritis/intestinal metaplasia,atypicalhyperplasia, 20 cases), gastric cancer(20 cases) and normal gastric mucosa(20 cases) were obtained from gastroscopic biopsy or surgically removed.The quantitative real-time PCR, Western blot and immunohistochemical staining were performed for examining the m RNAs and proteins expressions of RUNX3, Jagged1,E-cadherin and N-cadherin in normal gastric mucosa, precancerous lesions and gastric cancer tissues. SPSS 19.0 and IPP statistical software packages were used for statistical analysis.Results: The m RNAs and proteins expressions of RUNX3 and E-cadherin was gradually lower in normal gastric mucosa, precancerous lesions and gastric cancer tissues.Comapared with that in the normal gastric mucosa, the expressions in precancerous lesions and gastric cancer tissues decreased significantly(P<0.05), and there were significantly differences between the precancerous lesions and gastric cancer tissues(P<0.05). However,the m RNAs and proteins expressions of Jagged1 and N-cadherin was gradually higher.Compared with the that in normal gastric mucosa tissues, the expressions in precancerous lesions and gastric cancer tissues increased significantly(P<0.05). And the same results were also observed in the immunohistochemical staining. Results of Spearman rank correlation analysis showed a positive correlation between the expressions of RUNX3 and E-cadherin(r=0.397, P<0.05), and a negative correlation between the expressions of RUNX3 and Jagged1, N-cadherin(r=-0.323, P<0.05; r=-0.435, P<0.05).Conclusion: Expressions of RUNX3 and E-cadherin showed decreases or deletions in the development progress from normal gastric mucosa, precancerous lesions to gastriccancer tissues. However, the contrasts were with Jagged1 and N-cadherin.There were a positive correlation between the expressions of RUNX3 and E-cadherin and a negative correlation between the expressions of RUNX3 and Jagged1, N-cadherin. These results indicate that the expression levels of RUNX3, Jagged1, E-cadherin and N-cadherin in gastric mucosa tissues have correlations with the tissue malignancy levels. The RUNX3,Jagged1, E-cadherin and N-cadherin may play inportant roles in the occurrence and development of gastric cancer. However, the mechamisms should be made futher study. |
