Objective: Through atmospheric vacuum airtight closed anoxia resistance experiments in mice resistance to anoxia experiment and nitrite poisoning experiment, to observe the anti- hypoxia of Oxytropis ochrocephala effect. Methods: Experiment were divided into normal pressure closed anoxia resistance experiment, reduced pressure closed anoxia resistance experiment and nitrite poisoning experiment three parts, each part 60 mice. Every part of these 60 mice were randomly divided into normal saline group, O. ochrocephala low dose group, O. ochrocephala middle dose group,O. ochrocephala high dose group,ginseng total saponins group(positive control) and propranolol group(positive control), each group of 10,The dosage of administration of O. ochrocephala are 1.5 g/kg,1.0 g/kg and 0.5 g/kg for high dose,middle dose and low dose,while 0.6 g/kg and 150 mg/kg are for ginseng total saponins group and propranolol group, The same volume of saline is given for blank control group. Each group is continuously fed for 7 days, In the last 1 h after lavage for normal pressure closed anoxia resistance experiment, reduced pressure closed anoxia resistance experiment and nitrite poisoning experiment. Record the survival time of mice, and the detection analysis of heart and brain SOD, MDA, GSH-Px, Bcl-2 and Bax. Results: Firstly, compared to the blank control group, O. ochrocephala high dose, ginseng total saponins and propranolol group mice were exposed to hypoxia time extended obviously in normal pressure closed anoxia experiment and sodium nitrite poisoning, the biochemical indexes were improved(P < 0.01). Secondly, compared to the blank control group, the activities of SOD and GSH-Px are enhanced obviously, the expression of Bcl-2 protein are significantly increased,the content of MDA are decreased obviously, the biochemical indexes were improved(P < 0.01), the expression of Bax protein are significantly decreased, the biochemical indexes were improved(P < 0.05) in heart and brain of O. ochrocephala high dose, ginseng total saponins and propranolol groups in normal pressure closed anoxia experiment.Thirdly, for nitrite poisoning experiment, compared to the blank control group, the activities of SOD and GSH-Px are improved obviously, the contents of Bax and MDA are dropped significantly, the biochemical indexes were improved(P < 0.01), the expression of Bcl-2 protein are significantly improved, the biochemical indexes were improved(P < 0.05) in heart and brain of O. ochrocephala high dose, ginseng total saponins and propranolol groups. The activities of SOD are improved in heart and brain of O. ochrocephala middle dose group, the biochemical indexes were improved(P < 0.05).Fourthly, as to reduced pressure closed anoxia resistance experiment, compared to the blank control group, the activities of SOD and GSH-Px are enhanced obviously, the contents of Bax and MDA are decreased significantly, the biochemical indexes were improved(P < 0.01), the expression of Bcl-2 protein are significantly increased, the biochemical indexes were improved(P < 0.05) in heart and brain of O. ochrocephala high dose, ginseng total saponins and propranolol groups. While in O. ochrocephala middle dose group, the activities of SOD and GSH-Px are improved in heart and brain, and the expression of Bax protein are decreased in brain, the biochemical indexes were improved(P < 0.05). Conclusion: The alcohol extraction of O. ochrocephala could significantly prolong the mice in the condition of normal pressure closed anoxia and sodium nitrite poisoning survival time, significantly improve the activities of SOD and GSH-Px and the expression of Bcl-2 protein in mice,and significantly decreased the expression of Bax protein and the content of MDA. So the alcohol extraction of O. ochrocephala could improved the hypoxia adaptation and have a certain effect of oxygen antagonism.
