Objective:Exploring the changes and clinical significance of cytokines(IFN-γ, IL-2, IL-12, IL-4, IL-6, IL-10, TNF-α, IL-27, IL-17, IL-11, TPO, TGF-β) in the same batch ITP patients.Methods:1. Collecting the plasma of 20 ITP patients before and after treatment and other 20 healthy controls(control group). Then detecting the plasma levels of different cytokines with ELISA. 2. Detecting the different groups’ cytokines’ m RNA level in peripheral mononuclear cell with Real-time RT-PCR.Statistic method: Data were analysed by SPSS17.0 statistical software. Normal distribution data were presented with mean ± standard deviation(`x±s) and the comparison between before treatment and treatment group were conducted with the paired t test, while the data between before treatment and control group were analysed with independent sample t test. Skewed distribution measurement data using rank sum test. test level take α = 0.05.Results:1. In compared to the contents of different cytokines of different group, there were no statistic differences in IFN-γ, TNF-α, IL-2, IL-12, IL-4, IL-6, IL-10, IL-27 between before treatment and control group. Furthermore, no statistic differences were found in contents of IFN-γ, TNF-α, IL-2, IL-12, IL-4, IL-6, IL-10, IL-27 between before treatment and treatment group. Besides IL-12p40 m RNA expression level of before treatment(4.97±3.62) were dramatically higher than control group(2.33±2.15), among these three groups of peripheral mononuclear cells, m RNA expression levels of IFN-γ, TNF-α, IL-2, IL-12p40, IL-4, IL-6, IL-10 and IL-27 had no statistic differences. 2. while the plasma levels of IL-17, TPO, IL-11 before treatment(12.29±8.63pg/ml, 133.21±61.34pg/ml, 564.21±893.77pg/ml) were much higher than control group(6.89±4.70 pg /ml, 46.08±41.71 pg /ml, 52.98±1.93 pg/ml)(P<0.01). In compared to the plasma level of IL-11 has no statistic differences between before treatment and after group. In contrast, the plasma levels of IL-17 and TPO were significantly decrease after treatment(8.20±4.43 pg /ml, 77.07±63.86 pg/ml)(P<0.05). 3. The plasma level of TGF-β before treatment(55.76±40.49pg/ml) were much higher than control group(435.65±227.42pg/ml)(P<0.01), while the plasma level of TGF-β after treatment(154.68±71.11pg/ml) were dramatically increase(P<0.01). Among these three groups of peripheral mononuclear cells, m RNA expression levels of TGF-β, IL-17, TPO and IL-11 had no statistic differences, respectively.Conclusion:TPO, IL-11, IL-17 and TGF-β these four cytokines play very important roles in pathogenesis of ITP, and the detection may have a relative significance in auxiliary diagnosis and estimation of prognosis.
