Objectives To study the expression levels of MMP-14 and TIMP-2 in tissues of thyroid papillary carcinoma and thyroid benign lesion, to analyse the relationship between expression levels of MMP-14 and TIMP-2 and clinicopathological features of thyroid papillary carcinoma. To analyse relevance between MMP-14 and TIMP-2. It provides theoretical evidences for pathogenesis of thyroid papillary carcinoma and can help to early diagnosis and treatments of thyroid papillary carcinoma.Methods We gathered 2 groups paraffin-embedded tissue samples in thyroidectomy of Tangshan Gongren Hospital from Jan 2013 to Jun in 2014, consist of 37 samples of metastatic carcinoma of lymph node, 31 samples of thyroid benign lesion, to compare the differences of positive expression rate among 2 groups by statiscal method. To analysis the relationship between expression levels of MMP-14 and TIMP-2 in thyroid papillary carcinoma samples and the clinicopathological features(such as sex, age, single or double, stove oven and above, lesion size, whether envelope invaded, and whether lymph node matastasis. To analysis relativity between expression levels of them in samples of thyroid papillary carcinoma.Results 1 The positive expression rate of MMP-14 in tissues of thyroid papillary carcinoma and thyroid benign lesion of lymph node was 81.1% and 25.8%. Through the statistical analysis, expression levels of MMP-14 was higher in group thyroid papillary carcinoma than group thyroid benign lesion, with statistically significant difference(χ2=31.869, P <0.05). The expression levels of MMP-14 in thyroid papillary carcinoma has no relevance with the clinicopathological features of sex, age and focus number. the expression levels of MMP-14 in thyroid papillary carcinoma has high relevance with the clinicopathological features of lesion size, whether envelope invaded, and whether lymph node matastasis. MMP-14 was overexpression in thyroid papillary carcinoma(more than to 1 cm in diameter) than carcinoma less than or equal 1 cm(93.9% vs. 73.7%), with statistically significant difference(χ2=5.597, P <0.05). Expression levels of MMP-14 was higher in group with envelope invaded than group without envelope invaded(95.7% vs. 76.1%), with statistically significant difference(χ2=4.264, P <0.05). Expression levels of MMP-14 was higher in group with lymph node matastasis than group without lymph node matastasis(97.3% vs. 69.9%), with statistically significant difference(χ2=10.744, P <0.05). 2 The positive expression rate of TIMP-2 in tissues of thyroid papillary carcinoma and thyroid benign lesion of lymph node was 77.8% and 19.3%. Through the statisticalanalysis, expression levels of TIMP-2 was higher in group thyroid papillary carcinoma than group thyroid benign lesion, with statistically significant difference(χ2=33.692, P <0.05). The expression levels of TIMP-2 in thyroid papillary carcinoma has no relevance with the clinicopathological features of sex, age and focus number. the expression levels of TIMP-2 in thyroid papillary carcinoma has high relevance with the clinicopathological features of lesion size, whether envelope invaded, and whether lymph node matastasis. Expression levels of TIMP-2 was higher in group without envelope invaded than group with envelope invaded(83.4% vs. 60.9%), with statistically significant difference(χ2=5.110, P <0.05). Expression levels of TIMP-2 was higher in group without lymph node matastasis than group with lymph node matastasis(88.7% vs. 62.7%), with statistically significant difference(χ2=8.864, P <0.05). 3 To analyse relevance among expression of MMP-14 and TIMP-2. Spearman’s Rank Correlation Analysis was used to examine the significance. Expression levels of MMP-14 and TIMP-2 are negative correlated with thyroid papillary(P <0.01).Conclusions The expression levels of MMP-14 and TIMP-2 in thyroid papillary carcinoma has high relevance, It means MMP-14 and TIMP-2 may play a significant role in the initiation and progression of thyroid papillary carcinoma. Expression levels of MMP-14 has negative correlation with expression levels of TIMP-2,it suggests that there may be inner link. the expression levels of MMP-14 in thyroid papillary carcinoma has high relevance with the clinicopathological features of lesion size, whether envelope invaded, and whether lymph node matastasis. the expression levels of TIMP-2 in thyroid papillary carcinoma has high relevance with the clinicopathological features of lesion size, whether envelope invaded, and whether lymph node matastasis. Expression levels of MMP-14 has positive negative with expression levels of TIMP-2. It means MMP-14 and TIMP-2 may play a synergistic role in the initiation and progression of thyroid papillary carcinoma.
